摘要
目的探讨环状RNA MYLK在卵巢癌细胞SKOV3的增殖、侵袭及小管形成中的调节作用。方法pcDNA-MYLK过表达质粒转染SKOV3细胞。RT-PCR检测RNA表达;克隆形成和EdU染色检测增殖;Transwell检测侵袭;小管形成实验检测体外血管生成情况;Western印迹检测蛋A表达。结果与对照组比较,MYLK过表达显著增加克隆形成率(P<0.05);MYLK过表达显著增加EdU阳性细胞率,显著上调Ki67蛋白表达,显著下调Bax/Bd-2表达比值(P<0.05);MYLK过表达显著增加侵袭细胞数量和体外血管形成结节数量(P<0.05);MYLK过表达在mRNA和蛋白水平均显著上调血管内皮生长因子(vascular endothelial growth factor,VEGF)和波形蛋白(Vimentin)的表达,显著下调上皮钙黏蛋白(E-cadherin)的表达(P<0.05)。结论环状RNA MYLK过表达可诱导卵巢癌SKOV3细胞增殖、侵袭及体外血管形成。
Objective To investigate the regulatory effect of circmas MYLK on SK0V3 proliferation,invasion and tubule formation in ovarian cancer cells.Methods SK0V3 cells were transfected with pcDNA-MYLK overexpression plasmid.RNA expression was detected by RT-qPCR.Clonal formation and EdU staining were used to detect proliferation.Transwell was used to detect invasion.Tubule formation assay was used to detect angiogenesis in vitro.Western blotting was used to detect protein expression.Results Compared with the control group,MYLK overexpression significantly increased the clonal formation rate(P<0.05).MYLK overexpression significantly increased the EdU positive cell rate,significantly upregulated the expression of Ki67 protein,and significantly down-regulated the expression ratio of Bax/Bcl-2(all P<0.05).MYLK overexpression significantly increased the number of invading cells and the number of angiogenic nodules in vitro(P<0.05).MYLK overexpression significantly up-regulated VEGF and Vimentin expression at both mRNA and protein levels,and down-regulated E-cadherin expression significantly(P<0.05).Conclusion Overexpression of circrnas MYLK can induce proliferation,invasion and in vitro angiogenesis of ovarian cancer SKOV3 cells.
