摘要
金花葵根部含有丰富得黄酮类物质,黄酮有抗肿瘤、抗氧化、抗衰老等特性,具有较高的医疗保健价值。通过Box-Behnken响应面法优化超高压提取金花葵根部黄酮的工艺条件,同时探究提取工艺对黄酮提取物抗氧化性的影响。根据超高压压力、保压时间、乙醇体积分数三个单因素试验,进行三因素三水平的响应面分析。响应面优化表明,在超高压压力400MPa、保压时间8.3min、乙醇体积分数70%优化条件下,黄酮得率为(3.87±0.13)%,模拟值与实验测定基本相同。超高压提取的黄酮提取物对DPPH自由基和羟基自由基的清除作用的IC50分别为2.71mg/mL和5.34mg/mL,优于加热回流法和超声辅助法。说明超高压提取法既能保持较高的黄酮得率,并且黄酮提取物有良好的抗氧化性。
Aurea helianthustems root is rich in flavonoids. Flavonoids have anti-tumor,anti-oxidation,anti-aging properties,and have high medical care value. Box-Behnken response surface methodology was used to optimize the ultra-high pressure extraction process of flavonoids in the Roots of Aurea helianthustems. The effect of extraction process on antioxidant properties of flavonoid extract was also studied. Response surface analysis of three factors and three levels was carried out to optimize the ultra-high pressure extraction process based on ultra-high pressure,dwell time and ethanol volume fraction three single factor tests. The results showed that the optimum extraction conditions were :ultra-high pressure 400 MPa,dwell time 8.3 min,ethanol volume fraction 70%. The yield of flavonoids under the above conditions was(3.87±0.13)%. The simulated value is very close to the experimental value. The IC50 of scavenging capacity of the flavonoid extract obtained by the ultra-high pressure method on DPPH and·OH were 2.71 mg/mL and 5.34 mg/mL. The ultra-high pressure method is superior to the heated reflux method and the ultra-sonic assisted method.This showed that the ultra-high pressure extraction method can maintain a high yield of flavonoids and keep good antioxidant properties of the flavonoid extract.
