摘要
目的通过小鼠卵巢体外培养过程添加促卵泡刺激素(FSH)保护性干预,观察体外培养过程中FSH对血管内皮生长因子(VEGF)及血管内皮生长因子受体-2(VEGFR-2)表达的影响。方法将9只4周龄C57BL/6J雌性小鼠卵巢组织随机分为新鲜对照组(CG组)、体外培养对照组(VCG组)、0.3 IU/mL FSH干预体外培养组(IG-FSH组),通过免疫组织化学技术及实时荧光定量PCR技术观察并分析各组卵巢组织VEGF及VEGFR-2表达情况。结果IG-FSH组VEGF、VEGFR-2蛋白及mRNA表达量高于VCG组及CG组,差异均有统计学意义(P<0.05)。IG-FSH组及CG组VEGF mRNA表达量高于VCG组(P<0.05),但IG-FSH组及CG组比较差异无统计学意义(P>0.05)。IG-FSH组VEGFR-2 mRNA表达量高于CG组和VCG组(P<0.05)。结论FSH体外干预可有效促进VEGF及VEGFR-2的表达,在小鼠卵巢组织体外培养过程中发挥积极作用。
Objective To investigate the effect of follicle-stimulating hormone(FSH)on the expression of vascular endothelial growth factor(VEGF)and vascular endothelial growth factor receptor-2(VEGFR-2)in the process of mouse ovary culture in vitro by adding FSH protective intervention.Methods The ovarian tissues of 94-week-old C57BL/6J female mice were randomly divided into fresh control group(CG group),culture in vitro control group(VCG group),0.3 IU/mL FSH intervention culture in vitro group(IG-FSH group).The expression levels of VEGF and VEGFR-2 of ovarian tissues in each group were observed and analyzed by immunohistochemistry and real-time fluorescence quantitative PCR technology.Results The expression levels of VEGF,VEGFR-2 protein in the IG-FSH group were significantly higher than those in the VCG group and the CG group,the differences were statistically significant(P<0.05).VEGF mRNA expression levels in IG-FSH group and CG group were higher than those in VCG group(P<0.05),but there were no significant difference between IG-FSH group and CG group(P>0.05);VEGFR-2 expression level in IG-FSH group was higher than those in CG group and VCG group(P<0.05).Conclusion FSH intervention in vitro can effectively promote the expression of VEGF and VEGFR-2,and plays an active role in the process of mouse ovary tissue culture in vitro.
