摘要
目的:观察黄地安消胶囊对2型糖尿病模型(T2DM)GK大鼠胰岛素耐量(ITT)及胰岛素受体底物-1(IRS-1)表达的影响。方法:选取模型成功的GK大鼠纳入实验,随机分为模型组,参芪降糖颗粒组,黄地安消胶囊高、中、低剂量组,连续灌胃给药6周,测定各组大鼠给药6周后的胰岛素耐量试验,观察其对血糖的影响。HE染色观察大鼠胰腺组织病理损伤程度;RT-qPCR技术检测骨骼肌IRS-1 mRNA表达水平,Western bolt检测骨骼肌IRS-1蛋白的表达水平。结果:与模型组相比,黄地安消胶囊剂量组血糖水平明显降低(P<0.05),胰腺组织病理损伤程度具有不同程度的减轻;骨骼肌组织中IRS-1 mRNA和蛋白表达水平显著增加(P<0.05)。结论:黄地安消胶囊对T2DM模型GK大鼠血糖具有一定的改善作用,其机制可能与增强IRS-1蛋白表达,促进葡萄糖消耗有关。
Objective:To observe the effect of Huangdi Anxiao Capsule on insulin tolerance(ITT)and insulin receptor substrate-1(IRS-1)expression in GK rats with type 2 diabetes mellitus(T2DM).Methods:The successful GK rats were randomly divided into model group,Shenqi Jiangtang granule group and Huangdi Anxiao capsule high,medium and low dose groups.Rats in each group were administered with continuous intragastric administration for 6 weeks.The insulin tolerance test after 6 weeks of the drug was observed to observe its effect on blood sugar.The pathological damage of pancreatic tissue was observed by HE staining.The expression of IRS-1 mRNA in skeletal muscle was detected by RTqPCR and the expression of IRS-1 protein in skeletal muscle was detected by Western blot.Results:Compared with the model group,the blood glucose level of Huangdi Anxiao capsule group was significantly decreased(P<0.05),the degree of pathological damage of pancreatic tissue was reduced to some extent;the expression level of IRS-1 mRNA and protein in skeletal muscle tissue was significantly increased(P<0.05).Conclusion:Huangdi Anxiao capsule can improve the blood glucose of T2DM model GK rats,and its mechanism may be related to improving insulin resistance and enhancing IRS-1 protein expression.
