摘要
目的探讨不同浓度β-羟基丁酸(beta-hydroxybutyrate,BHB)对人星形胶质瘤U251细胞增殖及磷脂酶D1(phospholipase D1,PLD1)表达的影响。方法体外培养人星形胶质瘤U251细胞,用0、2 mmol/L、4 mmol/L、8 mmol/L、10 mmol/L BHB处理24 h,采用MTT法检测各组细胞的活力;用Western blot实验检测各组细胞的PLD1蛋白表达水平。结果2 mmol/L、4 mmol/L、8 mmol/L、10 mmol/L BHB组U251细胞的活力均被明显抑制(均P<0.05),并随着BHB浓度的增高,细胞活力抑制越明显。2 mmol/L、4 mmol/L、8 mmol/L、10 mmol/L BHB组U251细胞的PLD1蛋白表达水平均明显下降(均P<0.05),并随着BHB浓度的增高,PLD1蛋白表达水平逐渐下降。结论BHB具有抑制人星形胶质瘤U251细胞增殖的作用,其机制可能与下调PLD1表达有关。
Objective To investigate the effects of different concentrations of beta-hydroxybutyrate(BHB)on its proliferation and phospholipase D1(PLD1)expression of human astroglioma cell line U251.Methods Primary cultured human astroglioma U251 cells were pretreated with BHB at various concentrations(2 mM,4 mM,8 mM and 10 mM)for 24 hours.The cell viability was detected by MTT assay.The expression of PLD1 protein was detected by Western blot.Results BHB concentrations of 2 mM,4 mM,8 mM and 10 mM could significantly inhibit the activity of U251 cells(P<0.05),the inhibition of cell activity was more obvious with the increase of BHB concentration.BHB concentrations of 2 mM,4 mM,8mM and 10 mM could significantly inhibit PLD1 protein expression level of U251 cells(P<0.05),and decrease with the increase of BHB concentration.Conclusion BHB can inhibit the proliferation of glioma U251,and its mechanism may be related to the down-regulation of PLD1 expression.
