陈皮特征图谱方法比较研究

Comparative study on characteristic chromatogram of Citri Reticulatae Pericarpium

目的:建立陈皮特征图谱,并比较探讨相对保留时间法、双标线性校正法及随行对照法在陈皮特征图谱建立中的优缺点。方法:采用十八烷基键合硅胶为填充剂的液相色谱柱,以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱。分别采用相对保留时间法、双标线性校正法及随行对照法建立陈皮特征图谱,并比较分析其各自的优缺点。结果:20批陈皮样品液相色谱图中选定6个共有峰建立陈皮的特征图谱,其中相对保留时间法所需对照品最少,但受色谱柱影响最大,色谱峰定性准确度较差;双标线性校正法采用2个对照品对其他峰进行校正,色谱峰定性准确度高;随行对照法采用对照药材及对照品双重定性色谱峰,色谱峰定性准确度较高,但需较多标准物质。结论:建立的陈皮特征图谱可以用于陈皮的质量评价。双标线性保留时间校正法是陈皮特征图谱建立的较优方法。

Objective:To establish a characteristic chromatogram of Citri Reticulatae Pericarpium and compare and discuss the advantages and disadvantages of relative retention time method,linear calibration using two reference substances and accompanying control method in the establishment of characteristic chromatogram of Citri Reticulatae Pericarpium.Methods:A liquid chromatography column using octadecyl bonded silica gel was adopted as the solid phase,with acetonitrile-0.1%phosphoric acid as the mobile phase in gradient elution.The relative retention time method,linear calibration using two reference substances and accompanying control method were used to establish the characteristic chromatogram of Citri Reticulatae Pericarpium and their respective advantages and disadvantages were compared and analyzed.Results:A total of 6 common peaks in 20 batches of samples were selected to establish the characteristic chromatogram of Citri Reticulatae Pericarpium.Among them,the relative retention time method required the least reference substance,but was most affected by the column,and the chromatographic peak qualitative accuracy was poor.Linear calibration using two reference substances used two reference substances to calibrate other peaks,and the chromatographic peak qualitative accuracy was high.The accompanying control method used the dual qualitative chromatographic peaks of the control medicinal material and the reference substance,and the chromatographic peak qualitative accuracy was high.Conclusion:The characteristic chromatogram of Citri Reticulatae Pericarpium can be used to evaluate the quality of Citri Reticulatae Pericarpium.Linear calibration using two reference substances is the superior method for establishing the characteristic chromatogram.
