大鼠“体外延长尾静脉”采血法的构建与评价 被引量:1

摘要 探讨一种符合3R实验动物伦理原则的大鼠微创采血方法。自制大鼠固定装置和体外延长尾静脉采血装置,测量不同尾部浸泡温度的采血成功率、最大采血量和1 ml血量的采血时间。结果表明,(42±2)℃温水中浸泡有利于大体重大鼠尾部取血;(52±2)℃会烫伤大鼠尾部。10周龄大鼠平均采血量为(1.52±0.6)ml,28周龄大鼠平均采血量为(2.3±1.0)ml,1 ml血量的采集时间在1 min左右完成。结论:采用自制采血装置可以延长体外尾静脉,血液无污染,可多次成功采血,在大鼠安全范围内采集足量血液,满足检测的需要,对动物伤害小,体现了3R原则。 A minimally invasive method for blood collection in experimental rats was proposed according to the ethical principles of 3R experimental animals.Self-made rat fixation device and in vitro extended tail vein blood collection device was completed.The success rate of blood collection at different tail soaking temperatures,the maximum blood collection amount and the collection time of 1 ml blood volume were measured.The study found that immersion in warm water at(42±2)℃was most beneficial to the extraction of blood from the tail of rats.(52±2)℃will burn the rat tail.The average blood collection volume of rats in 10 weeks old was(1.52±0.6)ml,and the average blood collection volume of rats in 28 weeks old was(2.3±1.0)ml.The collection time of 1ml blood volume was completed in about 1min.The self-made blood collection device can realize the extension of the tail vein in vitro,to collect blood without pollution and successfully collect blood repeatedly.Extending the tail vein in vitro can collect a sufficient amount of blood in the safe range of the rat to meet the detection needs,and the damage to the animal is small.This reflects the 3R principle.
