基于组学数据的藜麦油脂储存蛋白的生物信息学分析 被引量:1

摘要 采用同源对比方法,从藜麦的基因组中挖掘油脂储存蛋白序列,并利用生物信息学软件和转录组数据对挖掘的基因蛋白序列的理化性质、二级结构、亚细胞定位、进化树、组织表达情况等进行了分析。在藜麦基因组中确定了15个基因与油脂储存密切相关。15个油脂储存基因分布在9条染色体上,其氨基酸数目为153~675个,等电点为5.7~10.1。藜麦油脂储存蛋白的二级结构以无规则卷曲和α-螺旋为主。利用MEGA软件进行进化树分析,可将藜麦的油脂储存蛋白分为6个分枝,且基因表达情况与分枝情况具有相似性。藜麦油脂储存蛋白基因具有保守的理化特性,其表达具有组织特异性,且大多数成员在种子中的表达量显著增加。 The genes related to oil storage were identified from the genome of quinoa by blast method,and the physical and chemical properties of protein,secondary structure,subcellular location,phylogenetic tree and tissue expression were analyzed by bioinformatics software and transcriptome data.Bioinformatics analysis indicated that 15 oil storage genes maps on 9 chromosomes.The amino acids and isoelectric points were 153~675 and 5.6~10.1,respectively.Secondary structure prediction results showed that random coil and alpha helix were common structures.Phylogenetic tree was constructed by MEGA software.It showed that 15 proteins divided into six branches.The gene expression was consistent with relationship,which indicated that the genetic relationship and gene function were related.Quinoa storage oil protein gene family members contain conservative gene structure.Their expressions are tissue-specific,and most of the members are up-regulated in seed period.
