盐杜仲标准汤剂制备及指纹图谱研究 被引量:2

摘要 目的制备盐杜仲标准汤剂并建立质量标准。方法制备15批盐杜仲标准汤剂,以松脂醇二葡萄糖苷为指标成分,计算出膏率、指标成分含量和转移率,并建立超高效液相色谱指纹图谱分析方法,使用《中药色谱指纹图谱相似度评价系统》进行相似度评价,采用超高效液相色谱-四级杆-飞行时间-串联质谱技术对共有峰进行结构鉴定。结果15批盐杜仲标准汤剂中松脂醇二葡萄糖苷转移率为42.74%~71.43%,平均为57.26%。15批盐杜仲标准汤剂指纹图谱与对照指纹图谱相似度均大于0.95。指纹图谱共有峰9个,确认分别为梓醇、杜仲醇苷、京尼平苷酸、车叶草苷酸、绿原酸、olivil-4’-O-β-D-glucopyranoside、京尼平、松脂醇二葡萄糖苷和紫云英苷。结论本研究建立的盐杜仲标准汤剂制备工艺稳定,指纹图谱方法精密度、稳定性及重复性良好,可为盐杜仲配方颗粒的质量控制提供参考。 Objective To prepare standard decoction of salt Eucommiae Cortex;To establish the standards of quality control.Methods Totally 15 batches of standard decoction of salt Eucommiae Cortex were prepared.With pinoresinoldiglucoside as the detection index,the extraction rate,content and transfer rate of index components were calculated and the UPLC characteristic chromatograms was established.Similarity Evaluation System for Chromatographic Fingerprint of TCM was used to analyze and compare the characteristic chromatograms.The chemical structures of common peaks were identified with UPLC-Q-TOF-MS/MS in the standard decoction of salt Eucommiae Cortex.Results The extraction ratio of pinoresinoldiglucoside was from 42.74%to 71.43%,with an average value of 57.26%.The similarity between the fingerprint of standard decoction of salt Eucommiae Cortex pieces and the reference fingerprint was over 0.95.There were 9 peaks in the fingerprint,and the peaks were identified as catalpol,eucommioside,geniposidic acid,asperulosidic acid,chlorogenic acid,olivil-4’-O-β-D-glucopyranoside,genipin,pinoresinoldiglucoside,and astragalin.Conclusion The preparation method of standard decoction of salt Eucommiae Cortex established in this study is stable.The fingerprint method has good precision,stability and repeatability,which can provide reference for the quality control of salt Eucommiae Cortex formula granules.
