CRMP5 expression in gastric cancer cells and its effects on cell proliferation,migration and invasion

目的:研究胃癌细胞中坍塌反应调节蛋白5(CRMP5)的表达,并探讨CRMP5对胃癌细胞增殖、迁移、侵袭的影响及其作用机制。方法:采用qRT-PCR与Western blot法分别检测人胃黏膜上皮正常细胞(GES-1细胞)和胃癌细胞(AGS、MGC-803细胞)中CRMP5的表达水平。将si-con和si-CRMP5转染MGC-803细胞后,用MTT法检测细胞活力,双染法检测细胞凋亡率,Transwell法检测细胞迁移和侵袭能力,Western blot法检测细胞中CRMP5的表达及相关蛋白Ki-67、Cleaved Caspase-3、MMP-2和MMP-9的表达情况;均以未处理细胞作对照。结果:与胃黏膜上皮正常细胞相比,胃癌细胞中CRMP5的表达水平升高(P<0.05)。沉默CRMP5后,MGC-803细胞增殖能力减弱(P<0.05),凋亡率增加(P<0.05),细胞的迁移和侵袭能力减弱(P<0.05),且MGC-803细胞中Ki-67、MMP-2和MMP-9蛋白表达水平降低(P<0.05),Cleaved Caspase-3蛋白表达水平升高(P<0.05)。结论:CRMP5在胃癌细胞中高表达,干扰其表达可以明显抑制细胞增殖、迁移和侵袭。

Aim:To study the expression of collapsin response mediator protein 5(CRMP5)in gastric cancer cells,and investigate the effects of CRMP5 on proliferation,migration and invasion of gastric cancer cells and the mechanism.Methods:The expression levels of CRMP5 in gastric cancer cells(AGS and MGC-803)and normal gastric epithelial cells(GES-1)were detected using qRT-PCR and Western blot.The MGC-803 cells were transfected with si-con and si-CRMP5,respectively.MTT assay was used to detect cell viability.The apoptosis of cells was examined by Annexin V-FITC/PI double staining.Transwell was used to detect cell migration and invasion.The expression levels of CRMP5,Ki-67,Cleaved Caspase-3,MMP-2 and MMP-9 in the cells of each group were tested by qRT-PCR and Western blot.Results:The expression of CRMP5 in gastric cancer cells was significantly higher than those of normal gastric epithelial cells.After transfection of si-CRMP5,the proliferation of MGC-803 cell was weakened and apoptosis was enhanced,the migration and invasion of cells were decreased(P<0.05).The expression levels of Ki-67,MMP-2 and MMP-9 were decreased significantly,while the expression level of Cleaved Caspase-3 was increased significantly(P<0.05).Conclusion:CRMP5 is highly expressed in gastric cancer cells;interfering with its expression can significantly inhibit cell proliferation,migration and invasion.
