摘要
目的研究五子衍宗方对秀丽线虫生命特征的影响,对卵巢功能受损秀丽线虫生殖能力的保护作用及其作用机制。方法将秀丽线虫分为空白组、五子衍宗方提取液40 mg·mL^-1低剂量组、100 mg·mL^-1高剂量组,通过对秀丽线虫生命特征、寿命、身体弯曲频率、头部摆动频率、体长体宽、子代数的观察,研究五子衍宗方提取液的安全性;同时采用雷公藤甲素造模,形成卵巢功能受损秀丽线虫模型,观察五子衍宗方提取液对秀丽线虫子代数的影响,并采用RT-PCR检测ced-3和ced-9 mRNA的表达水平。结果与空白组比较,五子衍宗方两个剂量均能延长秀丽线虫寿命(P<0.05),且能提高秀丽线虫的生殖能力(P<0.05),对其他生命特征没有影响。100 mg·mL^-1剂量组对卵巢功能受损秀丽线虫的生殖能力有明显的保护作用(P<0.05),可使ced-3 mRNA表达水平下调,ced-9 mRNA表达水平上调。结论五子衍宗方在不影响秀丽线虫生命特征的前提下,能明显改善卵巢功能受损秀丽线虫的生殖能力。
Objective To determine the effect of Wuzi Yanzong formula (WZYZF) on the life characteristics of Caenorhabditis elegans (C.elegans) and the protective effect on the reproductive capacity of damaged ovarian nematodes,and the mechanism of action.Methods C.elegans were divided into a blank group,a low dose group (WZYZF extract:40 mg·mL^-1) and a high dose (WZYZF extract:100 mg·mL^-1) group.The safety of the extract was studied by observing the life characteristics of C.elegans:lifespan,body bending frequency,head swing frequency,body length and body width,and nematode generation.At the same time,triptolide was used to establish a nematode model with decreased ovarian function.The effect of WZYZF extract on the number of offsprings of model nematodes was observed,and the expression levels of ced-3 and ced-9 mRNA were detected by RT-PCR.Results Compared with the blank group,both the low and high dose WZYZF extract group prolonged the lifespan of nematodes (P<0.05),and improved the reproductive ability of C.elegans (P<0.05),but had no effect on other life characteristics.High-dose drugs obviously protected the reproductive ability of C.elegans with decreased ovarian function (P<0.05),and positively regulated the expression of ced-3 and ced-9 mRNA.Conclusion WZYZF formula can greatly improve the reproductive ability of damaged ovarian nematodes without effect on other life characteristics of C.elegans.
