维生素D3衍生物对激素依赖性皮炎豚鼠CD8^+杀伤性T细胞功能影响的机制 被引量:3

The mechanism of the effect of vitamin D3 derivatives on the function of CD8^+killer T cells in guinea pigs with hormone-dependent dermatitis

摘要 目的探讨维生素D3衍生物对激素依赖性皮炎豚鼠CD8^+杀伤性T细胞功能影响的机制。方法48只激素依赖性皮炎豚鼠按数字表法随机分为3组-模型组、他克莫司组与维生素D3组,每组各16只。他克莫司组与维生素D3组给予他克莫司与1,25-二羟维生素D3治疗,模型组不给予治疗,治疗观察时间为4周。评价3组豚鼠总体疗效,比较3组豚鼠治疗前、治疗后4周的皮肤油脂含量、血清白细胞介素-6(IL-6)与白细胞介素-8(IL-8)含量、CD8^+杀伤性T细胞比率。结果治疗后4周,模型组、他克莫司组与维生素D3组的总有效率为12.5%、75.0%和93.8%,差异有统计学意义(P<0.05)。维生素D3组与他克莫司组治疗后4周的皮肤油脂含量[(86.94±0.26)、(76.70±0.40)μg/cm^2]、CD8^+杀伤性T细胞比率[(28.14±2.18)%、(21.09±2.01)%]高于治疗前和模型组[(66.34±7.40)μg/cm^2、(13.22±1.48)%],且维生素D3组高于他克莫司组,差异均有统计学意义(P<0.05)。维生素D3组与他克莫司组治疗后4周的血清IL-6[(3.01±0.11)、(5.02±0.09)ng/L]与IL-8[(3.00±0.17)、(6.87±0.16)ng/L]含量低于治疗前和模型组[(9.17±0.14)、(11.67±0.84)ng/L],且维生素D3组低于他克莫司组,差异均有统计学意义(P<0.05)。结论维生素D3衍生物在激素依赖性皮炎豚鼠的应用能提高CD8^+杀伤性T细胞功能,抑制炎症因子的释放,有利于增加皮肤油脂含量,从而提高治疗效果。 Objective To explore the mechanism of the effects of vitamin D3 derivatives on the function of CD8^+killer T cells in guinea pigs with hormone-dependent dermatitis.Methods 48 cases guinea pigs with hormone-dependent dermatitis were equally randomly divided into three groups-model group,tacrolimus group and vitamin D3 group,each group 16 cases.The tacrolimus group and vitamin D3 group were treated with tacrolimus and 1,25-dihydroxyvitamin D3.The model group were not treated with treatment.The treatment observation time were 4 weeks.The overall curative effect of the 3 groups of guinea pigs was evaluated.The skin oil content,serum interleukin-6(IL-6)and interleukin-8(IL-8)levels,CD8^+killing ability of the 3 groups of guinea pigs before and 4 weeks after treatment The T cell ratio is compared.Results After 4 weeks treatment,the total effective rates of the model group,tacrolimus group and vitamin D3 group were 12.5%,75.0%and 93.8%,the difference was statistically significant(P<0.05).The skin oil content of the vitamin D3 group and the tacrolimus group after 4 weeks of treatment[(86.94±0.26),(76.70±0.40)μg/cm^2],CD8^+killer T cell ratio[(28.14±2.18)%,(21.09±2.01)%]were higher than before treatment and model group[(66.34±7.40)μg/cm^2,(13.22±1.48)%],and vitamin D3 group weres higher than the tacrolimus group,the differences were statistically significant(P<0.05).Serum levels of IL-6[(3.01±0.11),(5.02±0.09)ng/L]and IL-8[(3.00±0.17),(6.87±0.16)ng/L]in the vitamin D3 group and the tacrolimus group 4 weeks after treatment were lower than before treatment and model group[(9.17±0.14),(11.67±0.84)ng/L],and vitamin D3 group was lower than tacrolimus group,the differences were statistically significant(P<0.05).Conclusion The application of vitamin D3 derivatives in guinea pigs with hormone-dependent dermatitis can improve the function of CD8^+killer T cells,inhibit the release of inflammatory factors,and help increase skin oil content,thereby improve the therapeutic effect.
