慢性牙周炎患者唾液中miR-21水平与牙周指标、炎性细胞因子及蛋白酶相关分子的相关性研究 被引量:7

摘要 目的研究慢性牙周炎患者唾液中微小RNA(miR)-21水平与牙周指标、炎性细胞因子及蛋白酶相关分子水平的相关性。方法选择2016年3月至2019年3月该院收治的50例慢性牙周炎患者作为慢性牙周炎组,同期体检的50例健康志愿者作为健康对照组,检测唾液中miR-21、炎症细胞因子白细胞介素(IL)-1β、IL-6、IL-17、肿瘤坏死因子-α(TNF-α)、蛋白酶相关分子基质金属蛋白酶(MMP)-1、MMP-8、MMP组织抑制物(TIMP)-1水平及牙周指标菌斑指数(PLI)、牙龈指数(GI)、牙周探诊深度(PD)、附着丧失(AL)。结果慢性牙周炎组唾液中miR-21、IL-1β、IL-6、IL-17、TNF-α、MMP-1、MMP-8、TIMP-1水平及牙周指标PLI、GI、PD、AL均高于健康对照组,差异均有统计学意义(P<0.05);Pearson相关性分析显示,慢性牙周炎患者唾液中miR-21水平与IL-1β、IL-6、IL-17、TNF-α、MMP-1、MMP-8、TIMP-1、PLI、GI、PD、AL的水平呈正相关(P<0.05)。结论慢性牙周炎患者唾液中miR-21水平异常升高,且与牙周指标、炎性因子、蛋白酶分子水平存在一定的相关性,其可能参与了慢性牙周炎的发生、发展过程。 Objective To study the correlation between the levels of microRNA(miR)-21 in saliva and periodontal indexes,inflammatory cytokines,protease-related molecules in patients with chronic periodontitis.Methods A total of 50 cases of patients with chronic periodontitis admitted to the hospital from March 2016 to March 2019 were selected as the chronic periodontitis group and 50 cases of healthy volunteers who underwent physical examination at the same time were taken as the healthy control group.miR-21 in saliva,inflammatory cytokines interleukin(IL)-1β,IL-6,IL-17,tumor necrosis factor-α(TNF-α),protease-related molecules matrix metalloproteinase(MMP)-1,MMP-8 and tissue inhibitor of MMP-1(TIMP-1)and periodontal index plaque index(PLI),gingival index(GI),periodontal exploration depth(PD)and attachment loss(AL)were measured.Results The levels of miR-21,IL-1β,IL-6,IL-17,TNF-α,MMP-1,MMP-8,TIMP-1 in saliva and the levels of periodontal indexes PLI,GI,PD and AL in chronic periodontitis group were higher than those of healthy control group,and the differences were statistically significant(P<0.05).Pearson correlation analysis showed that the levels of miR-21 in saliva of patients with chronic periodontitis positively correlated with the levels of IL-1β,IL-6,IL-17,TNF-α,MMP-1,MMP-8,TIMP-1,PLI,GI,PD,AL(P<0.05).Conclusion The level of miR-21 in saliva of patients with periodontitis increase abnormally,and has a certain correlation with periodontal index,inflammatory factors,protease molecular level,which may be involved in the occurrence and development of chronic periodontitis.
