摘要
为明确灵芝病原真菌的归属,采用组织分离法[1]获得病原菌株,结合形态学及分子生物学方法对分离菌株进行鉴定,从显微特征和分子生物学对其进行分析鉴定;测定不同温度、pH、光照等条件下对病原菌产孢量、尖端菌丝数量的影响。结果表明:形态学鉴定及分子生物学鉴定为侧耳木霉,通过SPSS数据分析不同温度,pH对病原菌的产孢量与菌丝尖端数量有明显差异,光照则无明显差异。温度在15℃时产孢量为零,温度在15℃与30℃时尖端菌丝数量最少;pH为5与9时产孢量、尖端菌丝数量最少,因此灵芝病原菌为侧耳木霉,温度在15℃与30℃,pH为5与9时对产孢量和尖端菌丝数量影响最大。
In order to identify the ascription of Ganoderma lucidum pathogenic fungi,the pathogen strains were obtained by tissue isolation method[1].The isolated strains were identified by morphological and molecular biological methods.The effects of different temperature,pH and light on the sporulation and the number of hyphae were determined.The results showed that Trichoderma orientalis was identified as Trichoderma orientalis by morphology and molecular biology.SPSS data were used to analyze the difference of sporulation and hyphae tip number between different temperature and pH value,but there was no significant difference in light intensity.At 15℃,the spore production was zero,and the temperature was 15℃and 30℃,respectively.When the temperature was 15℃and 30℃,pH was 5 and 9,and the number of apical hyphae was the least.Therefore,the pathogen of Ganoderma lucidum was Trichoderma Pleurotus,the temperature of 15℃and 30℃,pH 5 and 9 had the greatest influence on the sporulation and the number of apical hyphae.
