摘要
目的探讨白虎汤不同相态对发热模型家兔的解热作用以及对家兔血清中TNF-α、IL-1β、IL-6水平的影响。方法将56只家兔随机分为空白组、模型组、经典白虎汤组、白虎汤纳米相态组、白虎汤真溶液组、白虎汤沉淀组、非那根组,每组8只。除空白组外每只家兔由耳缘静脉注入剂量为20μg/kg的脂多糖溶液,空白组注入等量生理盐水,制备家兔发热模型。各组在造模前0.5 h灌肠相应药物或生理盐水。给药5 h后,测量给药期间家兔的体温变化,并利用ELISA法检测各组家兔血清中炎性因子TNF-α、IL-1β、IL-6的水平。结果白虎汤纳米相态组对发热家兔一般状态的改善效果优于白虎汤其他相态组。测量各实验组家兔体温,非那根组可降低家兔的第一次发热体温。白虎汤各相态组均可有效降低家兔的第二次发热体温,纳米相态组的降温程度优于其他相态组。对家兔炎症因子进行测量,白虎汤各相态组的家兔血清中IL-1β、IL-6和TNF-α含量均低于模型组(P<0.01),白虎汤真溶液组和白虎汤沉淀组的家兔血清中IL-1β和TNF-α含量均低于模型组(P<0.01,P<0.05),且在白虎汤各相态组中纳米相态组的炎性因子含量最低[IL-1β、IL-6和TNF-α的含量分别为(9.55±0.36)、(11.96±0.18)和(55.66±1.3)pg/ml]。结论白虎汤纳米相态对家兔的解热作用以及血清中炎性因子TNF-α、IL-1β、IL-6水平的下降程度均优于其他相态,表明白虎汤纳米相态是白虎汤解热机制的关键。
Objective To investigate the antipyretic effect of different phases of Baihu decoction in fever model rabbits and the effect of different phases on the serum TNF-α,IL-1βand IL-6 level in fever model rabbits.Methods A total of 56 rabbits were randomly divided into 7 groups:blank group,model group,traditional Baihu decoction group,Baihu decoction nano phase group,Baihu decoction true solution group,Baihu decoction precipitation group and phenergan group,with 8 rabbits in each group.Except for the rabbits in the blank group,each rabbit was injected with lipopolysaccharide solution in the 20μg/kg dose through the auricular vein to create the rabbit fever model,while each rabbit in the blank group was injected with normal saline.Half an hour before the modeling,drugs or saline were given.5 hours after administration,body temperature changes in rabbits during administration were measured,and levels of inflammatory factors TNF-α,IL-1βand IL-6 were detected by ELISA.Results The effect of the nanometer phase configuration of Baihu decoction on the general state of febrile rabbits was better than that of other phase configurations.The body temperature measured for the rabbits in each group showed that phenergan decreased the first fever temperature,while all phase configurations of Baihu decoction effectively reduced the second fever temperature and the effect of nano phase configuration was better than that of other phase configurations.According to the measurement of inflammatory factors in rabbits,the levels of IL-1β,IL-6 and TNF-αin serum of the nanometer phase group of Baihu decoction were lower than those of the model group(P<0.01),and the levels of IL-1βand TNF-αin serum of the Baihu decoction group and the precipitation group were lower than those of the model group(P<0.01 and P<0.05).The contents of inflammatory factors of the nanometer phase group were the lowest in each phase of baihu decoction〔the levels of IL-1β,IL-6 and TNF-αwere(9.55±0.36),(11.96±0.18)and(55.66±1.3)pg/ml〕.Conclusion The antipyretic effect of Baihu decoction nanophase on rabbit and the decrease in levels of inflammatory factors TNF-α,IL-1βand IL-6 are better than those of other phase states,indicating that nanophase of Baihu decoction is the key to the antipyretic mechanism of Baihu decoction.
