摘要
目的以液质联用法(LC-MS)为标准,进行胶体金试纸法检测头发样本中吗啡(MOP)和甲基苯丙胺(MA)的准确性及可靠性研究。方法头发依次用甲醇、蒸馏水和甲醇进行清洗,烘干后剪碎,精密称取150 mg,甲醇37℃孵育24 h,离心5 min,取上层液0.5 ml用滤膜过滤后进行LC-MS检测,剩余液体氮气吹干,蒸馏水复溶,进行胶体金试纸法检测,该检测结果与LC-MS检测结果进行对比分析。结果胶体金试纸法检测头发样本中MOP的灵敏度为88.24%,特异度为100%,MA的灵敏度为60.61%,特异度为100%,胶体金试纸法和LC-MS法检测MOP的一致性很高,检测MA的一致性较高。结论胶体金试纸法检测头发毒品特异性很高,MOP检测的灵敏度高于MA,方法的准确性和可靠性较好。
Objective The liquid chromatography-mass spectrometry LC-MS method was used as the golden standard to explore the accuracy and reliability of colloidal gold lateral assay in detecting morphine(MOP)and methamphetamine(MA)in hair samples.Methods Hair samples were cleaned with methanol,distilled water and methanol successively,cut into pieces after dried.Then 150 mg was weighted and incubated with methanol at 37℃for 24 hours.After centrifugation for 5 min,0.5 ml of the supernatant liquor was filtered by filter membrane for LC-MS detection.The remaining supernatant solution was dried by nitrogen gas,dissolved in pure water,and used to detect MOP and MA by colloidal gold lateral assay.The results of colloidal gold lateral assay were compared with those of LC-MS method.Results The sensitivity and specificity of the colloidal gold lateral assay were 88.24%and 100%for the MOP detection and 60.61%and 100%for the MA detection,respectively,all in the hair samples.The consistency of the colloidal gold lateral assay with the LC-MS method was excellent for MOP detection and moderate for MA detection.Conclusion The specificity of this method was very high,the sensitivity for the MOP detection was higher than MA detection,and the accuracy and reliability were quite good.
