摘要
目的利用Catwalk步态分析系统结合Bederson改进神经缺损评分客观评价大脑中动脉闭塞(MCAO)大鼠步态参数及评价电针效应。方法将24只清洁级雄性SD大鼠随机分为正常组(无手术、无干预)8只、MCAO模型组(MCAO造模、无干预)8只、电针组(MCAO造模、治疗)8只。采用Zea Longa线栓法制作MCAO动物模型。电针组采用双侧风池穴电针干预,正常组及MCAO模型组不予任何干预。对各组大鼠进行Bederson改进神经缺损评分以及Catwalk步态参数采集。结果与正常组比较,MCAO模型组、电针组大鼠造模后Bederson改进神经缺损评分升高(P<0.05);与MCAO模型组比较,电针组干预后Bederson改进神经缺损评分降低(P<0.05)。与正常组比较,MCAO模型组、电针组大鼠造模后大鼠Catwalk步态参数改变(P<0.05),表现为大鼠四肢运动时间延长、运动速度变慢、步幅缩短、步长缩短、步态周期中站立时间延长、摆动时间延长、摆动速度减小、前后肢BOS增加,行走-站立模式改变;与MCAO模型组比较,电针组干预后Catwalk步态参数好转(P<0.05)。Bederson改进神经缺损评分与Catwalk步态参数有一致性。结论Catwalk步态分析系统结合Bederson改进神经缺损评分在识别异常步态、评价针刺效应方面存在一定优势。电针干预可改善MCAO大鼠Bederson改进神经缺损评分,促进肢体运动功能的恢复。
Objective To objectively assess gait parameters and evaluate electroacupuncture effect using Catwalk gait analysis system plus the modified Bederson neurological deficit scale in rats with middle cerebral artery occlusion(MCAO).Method Twenty four clean grade male SD rats were randomized to a normal group(no operation and intervention)of 8 rats,a MCAO model group(MCAO model making and no intervention)of 8 rats and an electroacupuncture group(MCAO model making and treatment)of 8 rats.Animal MCAO model was made using Zea Longa thread occlusion method.The electroacupuncture group received intervention by electroacupuncture at bilateral points Fengchi(GB20)and the normal and MCAO model groups,no intervention.The modified Bederson neurological deficit scale score was recorded and Catwalk gait parameters were collected in various groups of rats.Result Compared with the normal group,the modified Bederson neurological deficit scale score increased in the MCAO model and electroacupuncture groups of rats after model making(P<0.05).Compared with the MCAO model group,the modified Bederson neurological deficit scale score decreased in the electroacupuncture group after intervention(P<0.05).Compared with the normal group,Catwalk gait parameters changed and were manifested in long limb movement time,slow movement speed,short stride,short step length,long standing time in the duty cycle,long swing time,low swing speed,fore-hind limb BOS increase and walking-standing mode change in the MCAO model and electroacupuncture groups of rats after model making(P<0.05).Compared with the MCAO model group,Catwalk gait parameters took a turn for the better in the electroacupuncture group after intervention(P<0.05).The modified Bederson neurological deficit scale score and Catwalk gait parameters showed consistency.Conclusion Catwalk gait analysis system plus the modified Bederson neurological deficit scale has some advantage in identifying abnormal gait and evaluating acupuncture effect.Electroacupuncture intervention can decrease the modified Bederson neurological deficit scale score and promote the recovery of limb motor function in MCAO rats.
