云南省腾冲市牛、羊中无形体感染的分子流行病学调查 被引量:5

摘要 为了解云南腾冲市牛、羊中无形体病的流行情况,本研究选择该市11个乡镇为采样点,采集牛、羊血液样本,参照无形体16S rRNA基因设计引物,采用PCR方法检测牛、羊无形体流行情况。结果显示,从758份牛和羊的血样中共检出阳性样品255份,总阳性率33.64%。其中羊血样本427份,检出114份,阳性率26.69%,分布于8个乡镇;牛血样本331份,检出阳性141份,阳性率为42.60%,11个乡镇均有检出。对扩增的16S rRNA基因(约660 bp)片段测序后,对该基因序列经同源性和遗传进化分析,对无形体进行种类鉴定,结果显示,检出的阳性样品共涵盖7种无形体,其中羊阳性检出样品中包括嗜吞噬细胞无形体(10份)、山羊无形体(1份)、绵羊无形体(72份)、边缘无形体(1份)、牛无形体(5份)和血小板无形体(25份),共6个种类;牛检出阳性样品中包括嗜吞噬细胞无形体(9份)、中央无形体(5份)、绵羊无形体(6份)、边缘无形体(65份)、牛无形体(4份)和血小板无形体(52份),共6个种类。以上结果表明,腾冲市养殖的牛、羊中存在无形体病的广泛流行,且流行的无形体种类复杂多样,对家畜健康威胁较大,有些种类是对人致病的人兽共患无形体,需加强防范。本研究为腾冲市乃至云南边境地区无形体病的防治提供了科学依据。 In order to understand the prevalence of Anaplasmosis among cattle and goats in Tengchong City,Yunnan province,blood samples were collected from11 villages.A 660 bp of 16 S rRNA gene sequence of Anaplasmosis family was amplified by PCR and sequenced.The species of Anaplasmosis were identified by homology comparison and phylogenetic analysis.of 758 blood samples,255(33.64%)positive samples were detected,which cover 7 kinds of Anaplasmosis.114(26.69%)out of 427 goat blood samples were positive.Six types of Anaplasmosis,including A.phagocytophilum(10),A.capra(1),A.ovis(72),A.marginale(1),A.bovis(5)and A.platys(25),were found in 8 towns.141 out of 331(42.60%)cattle blood samples were positive.Six types of Anaplasmosis,including A.phagocytophilum(9),A.centrale(5),A.ovis(6),A.marginale(65),A.bovis(4),and A.platys(52),were found in all of the 11 villages and towns.The results demonstrated that there is a wide spread epidemic of Anaplasmosis in cattle and goats in Tengchong City,which poses a great threat to the health of livestock.Some of the Anaplasmosis are highly pathogenic,thus strengthening prevention is in need.This study provides a scientific basis for the prevention and treatment of Anaplasmosis in Tengchong City and border areas of Yunnan.
