新型多巴胺荧光比率传感器构建的研究 被引量:1

摘要 采用谷胱甘肽作稳定剂制备了荧光金簇(AuGSH),并将其与间苯二酚相结合构建了新型的荧光比率传感器用于检测多巴胺。检测原理为:多巴胺和间苯二酚在强碱性环境中,可生成位于455 nm处的蓝色荧光物质(DR),其荧光强度会随着多巴胺浓度的增加而增强;多巴胺对位于588 nm处的AuGSH的荧光基本没有影响。以AuGSH的荧光为参比信号,通过测定DR和AuGSH的荧光比率(F455/F588),可实现对多巴胺的准确测定。实验结果表明,F455/F588与多巴胺在1~10μmol/L和10~20μmol/L 2个浓度范围内呈线性相关,检出限为0.23μmol/L。此外,实验还对常见的潜在干扰物进行了检测,结果显示潜在干扰物不影响多巴胺的检测,说明该方法具有良好的选择性。 Fluorescent gold clusters(AuGSH)was prepared by using glutathione as stabilizer,and used to develop a new type of fluorescence ratio sensor with resorcinol for detecting dopamine.The detection principle was as follows:in strong alkaline environment,dopamine and resorcinol could generate fluorescence substance(DR)at 455 nm,and its fluorescence intensity would increase with the dopamine.However,dopamine had little effect on fluorescence of AuGSH at 588 nm.Therefore,accurate determination of dopamine could be achieved by determining the fluorescence ratio of DR and AuGSH(F455/F588),in which the AuGSH served as the reference signal.The experimental results showed that F455/F588 was linearly correlated with the dopamine concentration in the range of 1~10μmol/L and 10~20μmol/L and the detection limit was 0.23μmol/L.In addition,the common potential interferences were also detected,and the results showed that the chosen interferences did not affect the detection of dopamine,indicating that the method has good selectivity.
