蒙古扁桃油对肾纤维化大鼠的保护作用研究 被引量:5

Protective Effect of Amygdalus Mongolica Oil on the Kidneys in Rats with Renal Fibrosis

摘要 目的:探究蒙古扁桃油对肾纤维化大鼠肾脏的影响。方法:将60只SPF级雄性SD大鼠按体重随机分为6组:模型对照组、假手术对照组、盐酸贝那普利0.0015 g/kg组和蒙古扁桃油5.0 g/kg、10.0 g/kg、15.0 g/kg组。除假手术对照组外其余各组大鼠均采用单侧输尿管结扎术(UUO)造模,连续灌胃给药6 w。术后42 d处死大鼠,处死前采集大鼠眼眶静脉血样本,处死后采集大鼠腹主动脉血样本,摘取输尿管结扎侧肾组织均用于检测肾纤维化相关指标。结果:与正常对照组比较,模型对照组大鼠肾脏脏器指数、血清肌酐(serum creatinine,Scr)、尿素氮(blood urea nitrogen,BUN)、白蛋白(albumin,ALB)、丙二醛(malondialdehyde,MDA)、肾组织MDA和羟脯氨酸(hydroxyproline,HYP)含量显著升高,大鼠24 h尿量、血清超氧化物歧化酶(superoxide dismutase,SOD)和组织SOD活力显著降低(P<0.01)。与模型对照组比较,蒙古扁桃油5.0 g/kg组血清ALB含量、肾组织MDA水平明显降低,血清SOD活力明显升高(P<0.05);蒙古扁桃油10.0 g/kg组肾脏脏器指数、血清Scr、BUN、ALB、MDA以及肾组织MDA、HYP含量明显降低,血清SOD及肾组织SOD活力明显升高(P<0.05);蒙古扁桃油15 g/kg组血清ALB含量明显降低(P<0.05)。结论:蒙古扁桃油对肾纤维化大鼠肾脏具有保护作用。 Objective:To explore the effects of Amygdalus mongolica oil on the kidneys of rats with renal fibrosis.Methods:60 SPF male SD rats were randomly divided into 6 groups according to their body weights:the model group,the sham operation group,benazepril hydrochloride group(0.0015 g/kg)and different doses of Amygdalus mongolica oil groups(5.0 g/kg,10.0 g/kg,15.0 g/kg).Except for the sham opertaion group,all other rats were modeled by unilateral ureteral ligation(UUO),and were given intragastric administration for 6 weeks.The rats were sacrificed 42 days after the operation.The orbital venous blood samples were collected before the sacrifice,the blood samples from abdominal aorta of the rats were collected after the sacrifice,the kidney tissues from the ureteral ligation side were taken to detect the related indicators of renal fibrosis.Results:Compared with the sham operaion group,the levels of kidney organ index,serum creatinine(Scr),blood urea nitrogen(BUN),albumin(ALB),malondialdehyde(MDA),tissue MDA and hydroxyproline(HYP)were significantly increased in the model group,the 24 h urine output,the serum level of superoxide dismutase(SOD)and the tissue SOD activity were significantly decreased(P<0.01).Compared with the model group,the serum content of ALB and the tissue level of MDA in 5.0 g/kg Amygdalus mongolica oil group were significantly reduced,and the serum activity of SOD was significantly increased(P<0.05).The kidney organ index,the contents of serum Scr,BUN,ALB,MDA,and the tissue contents of MDA and HYP were significantly reduced in 10.0 g/kg Amygdalus mongolica oil group,while the activities of serum SOD and tissue SOD were significantly increased(P<0.05).The serum content of ALB in 15 g/kg Amygdalus mongolica oil group was significantly reduced(P<0.05).Conclusion:Amygdalus mongolica oil has a protective effect on the kidneys of rats with renal fibrosis.
