摘要
目的探讨N-乙酰基转移酶Naa10p对人食管鳞状细胞癌(ESCC)细胞增殖、迁移及侵袭能力的影响。方法质粒转染质粒分别构建Naa10p基因过表达组、干扰表达组及相应对照组,过表达(con-Naa10p)及其对照组(con-NC)按2μg质粒:4μl脂质体,干扰组(sh-Naa10p)及其对照组(sh-NC)按4μg质粒:8μl脂质体。采用qRT-PCR检测不同组中表达水平,MTT法、划痕实验及Transwell法分别检测细胞增殖、迁移及侵袭能力。结果Naa10p基因过表达组人食管鳞状细胞癌细胞增殖、迁移及侵袭能力低于对照组,Naa10p基因干扰组人食管鳞状细胞癌细胞增殖、迁移及侵袭能力高于对照组,差异均有统计学差异(均P<0.05)。结论Naa10p基因高表达能够抑制ESCC细胞增殖、迁移及侵袭能力。
Objective To analyze the effect of N-acetyltransferase Naa10p on proliferation,migration and invasion of human esophageal squamous carcinoma(ESCC)cells.Methods Naa10p gene overexpression group,interference expression group and corresponding control group were constructed respectively using transfectable plasmid.The overexpression group(con-Naa10p)was matched with the control group(con-NC)with the ratio of 2μg plasmids:4μl lipidosomes.The interference expression group(sh-Naa10p)was matched with the control group(sh-NC)with the ratio of 4μg plasmids:8μl lipidosomes.qRT-PCR was used to detect the expression level among different groups.MTT test,scratch test and Transwell test were used to detect the cell proliferation,migration and invasion ability,respectively.Results The proliferation,migration and invasion of human ESCC cells were lower in the Naa10p gene overexpression group than in the control group,while the proliferation,migration and invasion of human ESCC cells i were higher in the Naa10p gene interference group than in the control group.Conclusion High expression of Naa10p gene can inhibit proliferation,migration and invasion of ESCC cells.
