摘要
目的探究白藜芦醇对甲状腺癌SW579细胞增殖的影响及其作用机制。方法取对数生长期SW579细胞,分为20 mmol/L白藜芦醇组、40 mmol/L白藜芦醇组、60 mmol/L白藜芦醇组、80 mmol/L白藜芦醇组、100 mmol/L白藜芦醇组、对照组和空白组,白藜芦醇各组加入相应浓度白藜芦醇培养,对照组加入等体积二甲基亚砜(DMSO)培养,空白组常规培养,培养24 h后采用噻唑蓝(MTT)法检测各组细胞增殖情况。取对数生长期SW579细胞,分为100 mmol/L白藜芦醇组和对照组,采用流式细胞术检测2组SW579细胞周期,采用Western blotting技术检测2组SW579细胞中磷酸化蛋白激酶B(p-AKT)、磷酸化雷帕霉素靶蛋白(p-mTOR)以及双特异性蛋白磷酸酶(BCDC25B)表达水平。结果SW579细胞增殖与白藜芦醇浓度呈依赖性,白藜芦醇浓度越高,增殖率越低。100 mmol/L白藜芦醇组G0/G1期细胞比例与对照组比较差异无统计学意义(P>0.05),S期细胞比例明显高于对照组(P<0.05),G2/M期细胞比例明显低于对照组(P<0.05)。100 mmol/L白藜芦醇组p-AKT、p-mTOR和CDC25B相对表达量均明显低于对照组(P均<0.05)。结论白藜芦醇可能通过抑制p-AKT、p-mTOR及CDC25B蛋白的表达,从而使SW579细胞分裂增殖时停滞于S期,进而显著抑制甲状腺癌SW579细胞增殖。
Objective It is to explore the effect of resveratrol on the proliferation of thyroid cancer SW579 cells and its mechanism.Methods The SW579 cells at logarithmic growth phase were taken and divided into 20 mmol/L resveratrol group,40 mmol/L resveratrol group,60 mmol/L resveratrol group,80 mmol/L resveratrol group and 100 mmol/L resveratrol group,control group and blank group,each resveratrol group was cultured with corresponding concentration of resveratrol,the control group was cultured with equal volume of dimethyl sulfoxide(DMSO),the blank group was cultured routinely,all the groups were cultured for 24 h.The thiazole blue(MTT)method was used to detect cell proliferation in each group.The SW579 cells at logarithmic growth phase taken and divided into 100 mmol/L resveratrol group and control group.Flow cytometry was used to detect the cell cycle of SW579 in each group,and Western blotting was used to detect the expression of phosphorylated protein kinase B(p-AKT),phosphorylated rapamycin target protein(p-mTOR),and bispecific protein phosphatase(BCDC25B)in the SW579 cells in each group.Results The proliferation of SW579 cells was dependent on the concentration of resveratrol.The higher the concentration of resveratrol,the lower the proliferation rate.The ratio of G0/G1 phase cells in the 100 mmol/L resveratrol group was not significantly different from that in the control group(P>0.05),the ratio of S phase cells was significantly higher than that in the control group(P<0.05),the ratio of G2/M phase cells was significantly lower than that of the control group(P<0.05).The relative expression levels of p-AKT,p-mTOR and CDC25B in the 100 mmol/L resveratrol group were significantly lower than those in the control group(all P<0.05).Conclusion Resveratrol may inhibit the expression of p-AKT,p-mTOR and CDC25B protein,which let SW579 cell division and proliferation stagnate in S phase,and then significantly inhibit the proliferation of thyroid cancer SW579 cell.
