摘要
目的采用HPLC法测定含红曲产品中的洛伐他汀及洛伐他汀酸。方法采用RP-HPLC法,比较不同来源洛伐他汀酸的校正因子,以洛伐他汀为对照,建立与其开环产物洛伐他汀酸的校正因子的关系,同时定量测定洛伐他汀和洛伐他汀酸的含量。结果洛伐他汀酸相对于洛伐他汀的相对校正因子为1.003,在优化的实验条件下,3.43~343μg·mL^-1洛伐他汀与峰面积的线性关系良好(r=0.9999),回收率为93.80%~96.30%,RSD=1.00%。结论所用方法准确性高,适用于含红曲产品中洛伐他汀、洛伐他汀酸及总洛伐他汀的定量分析及质量评价。
OBJECTIVE To simultaneously determine Lovastatin and lovastatin acid in health food containing red koji by HPLC.METHODS Lovastatin acid control solutions prepared by different ways were compared where Lovastatin was used as the reference.Relative calibration factor of lovastatin acid to Lovastatin were calculated and contents of active constituents were calculated based on the relative calibration factors.RESULTS Relative correction factors of lovastatin acid with reference to Lovastatin was 1.003.A method was established for simultaneous quantification determination of lovastatin and lovastatin acid in health foods containing red koji.The calibration curves were linear in the range of 3.43-343μg·mL^-1 with correlation coefficient larger than 0.9999.The recovery was in the range of 93.80%-96.30% with RSD(n=6)of 1.0%.CONCIUSION The method is accurate and reliable,and suitable for the determination of Lovastatin and lovastatin acid in health foods containing red koji.
