摘要
本试验旨在研究河南省猪圆环病毒2型(Porcine circovirus type 2,PCV2)流行株的分子变异情况。病料样本来自河南省5个疑似患有断奶仔猪多系统衰竭综合征(Post-weaning multisystemic wasting syndrom,PWMS)的猪场。先利用PCR方法进行PCV2的检测,阳性样本进行PCV2全基因组的克隆与序列分析。5个猪场疑似病料样本的PCV2检测结果均为阳性,这5个PCV2毒株的全基因组序列长度均为1767 bp,同源性介于96.1%~99.9%,与河南省其它PCV2毒株基因组序列的同源性介于95.6%~99.8%。结果说明,5个毒株基因组序列的同源性较高,与河南省其它PCV2毒株基因组序列的同源性也较高,PCV2毒株间的亲缘关系近,PCV2病毒进化速度较慢。
The purpose of this experiment was to study the molecular variation of porcine circovirus type 2(PCV2)in some areas of Henan Province.The samples were collected from five pig farms with suspected post-weaning multisystemic wasting syndrome(PWMS).PCR was used to detect PCV2,and the whole genome of the positive samples was cloned and sequenced.PCV2 was positive in all five pig farms.The length of the whole genome of five PCV2 strains was 1767 bp,and their homology ranged from 96.1%to 99.9%,and 95.6%to 99.8%with other PCV2 strains in Henan Province.The results showed that the gene sequences of the five strains had high homology,and also had high homology with other PCV2 strains in Henan Province,and the phylogenetic relationship between PCV2 strains was very close,and the evolution speed of PCV2 virus was slow.
