摘要
α(1,2)岩藻糖转移酶基因1(α-1,2-Fucosyltransferase gene 1,FUT1)是调节大肠杆菌F18黏附受体表达的一个候选抗性基因。实验旨在分析迪庆藏猪FUT1基因编码蛋白序列,为探索该基因编码蛋白的功能、建立地方猪种抗病育种候选基因和相关遗传标记以及分子育种提供一定的理论依据。本实验通过生物信息学方法对迪庆藏猪FUT1基因序列和氨基酸序列进行理化性质和分子结构预测分析,并比较分析猪、牛、人等物种遗传距离。结果显示:迪庆藏猪FUT1基因全长3045 bp,存在4个错义突变;FUT1基因编码蛋白分子式为C4916H7548N1390O1353S46,是一种亲水性蛋白,无信号肽和跨膜结构,有92个潜在的磷酸化位点;组成FUT1蛋白的一级结构中,亮氨酸比例最高,酪氨酸最低;二级结构主要以α螺旋为主,具有结构分子活性;三级结构中其同源蛋白为c2hlhA,属于岩藻糖基转移酶家族成员。对比物种间FUT1基因序列,迪庆藏猪与滇南小耳猪亲缘关系近,与其他物种较远。
Alpha-(1,2)Fucosyltransferase gene(FUT1)is a candidate gene that regulates the expression of E.coli F18 adhesion receptor,affecting diarrhea and edema in piglets.The bioinformatics analysis of the protein encoded by FUT1 not only provides reference for exploring the function of the protein encoded by this gene,but also provides a theoretical basis for establishing candidate genes for breeding and breeding of local pig breeds and related genetic markers and molecular breeding.This experiment uses bioinformatics methods to predict the physical and chemical properties and molecular structure of Diqing Tibetan pig FUT1 gene sequence and amino acid sequence,and compare the genetic distances of pigs,cattle,humans and so on.The results showed that the Diqing Tibetan pig FUT1 gene was 3045 bp in length and there were 4 missense mutation.The FUT1 encoded protein molecular formula was C4916H7548N1390O1353S46.FUT1 protein is a hydrophilic protein,no signal peptide,no transmembrane structure and with 92 potential phosphorylation sites.In the primary structure of FUT1 protein,leucine ratio was the highest and tyrosine was the lowest.Its secondary structure is mainly composed ofα-helix and has structural molecular activity.In the tertiary structure,its homologous protein is c2hlhA,which belongs to the family of modulation fucosyltransferase.In the comparison of FUT1 gene sequences between different species,Diqing Tibetan pig was closely related to Yunnan small ear pig,but far related to other species.
