摘要
研究金属锌离子对LOX的活性、热稳定性和过氧化产率的影响。对LOX与锌离子的相互作用进行了光谱学分析(包括紫外、荧光和圆二色光谱)。结果显示,锌离子可使LOX的紫外吸收在一定浓度范围内增强,但对荧光和圆二色光谱影响甚微。表明锌离子只是微小地改变了LOX的构象,并未影响其二级结构。添加锌离子未改变LOX的热稳定性。锌离子对LOX催化氧化甘油三亚油酸酯的活性影响较大,使其产率从5%提高到16%。
The effects of Zn2+on the activity,thermal stability and peroxide yield of soybean lipoxygensas(LOX)were studied.The interaction between LOX and Zn^2+ was analyzed by spectroscopy,including UV,fluorescence and circular dichroism.The results showed that the UV absorption of LOX was enhanced by Zn^2+ in a certain concentration range,but it had little effect on fluorescence and circular dichroism,indicating that Zn^2+ only slightly changed the conformation of LOX and did not affect its secondary structure.The addition of Zn^2+ did not change the thermal stability of LOX,but had great influence on catalytic oxidation activity of glycerol trilinoleate,which increased its yield from 5% to 16%.
