摘要
以黄金百香果茎尖为试验材料,进行茎尖离体芽的诱导和培养,研究茎尖外植体消毒最佳方式与时间,初代、继代增殖及生根培养过程中适宜的激素种类及浓度。结果表明:最佳的消毒方式为75%的酒精溶液消毒30 s后再用0.1%HgCl2消毒10 min;最适初代培养基为MS+6-BA 2.0 mg/L+GA30.5 mg/L+NAA 0.1 mg/L,有利于丛生芽的诱导;最适继代增殖培养基为MS+6-BA 1.5 mg/L+NAA 0.1 mg/L,适宜芽的增殖且利于壮苗;最适生根培养基为1/2 MS+NAA 0.1 mg/L+IBA 0.1 mg/L,形成的苗健壮,生根率高达95%。
Shoot tips of passion fruit(Passiflora edulis)were induced and cultured in vitro on mediums,and the best time for disinfection,the suitable hormones and their concentrations in the culture,subculture for proliferation,and the rooting culture were determined.The results showed that the best disinfection method is to disinfect the shoot tips with 75%alcohol solution for 30 seconds and then 0.1%HgCl2 for 10 minutes,and that the best culture medium is MS+6-BA 2.0 mg/L+GA 0.5 mg/L+NAA 0.1 mg/L,which is propitious to inducing of cluster shoots.The optimum subculture medium is MS+6-BA 1.5 mg/L+NAA0.1 mg/L,which is suitable for shoot proliferation and plantlet growth.The best rooting medium is 1/2 MS+NAA 0.1 mg/L+IBA 0.1 mg/L.The resulting tissue culture plantlets under these optimum conditions grows robust and have a rooting rate of up to 95%.
