摘要
目的:观察卵巢癌患者外周血循环肿瘤细胞耗氧量和糖酵解情况。方法:收集87例卵巢癌患者,应用密度梯度离心联合免疫磁珠富集法筛选卵巢癌患者外周血中的循环肿瘤细胞,用Seahorse检测循环肿瘤细胞及癌旁细胞的耗氧量和糖酵解水平。结果:卵巢癌细胞株A2780糖酵解水平(56.96±1.55)mpH·min-1、循环肿瘤细胞PFTE-6糖酵解水平(61.67±5.13)mpH·min-1和循环肿瘤细胞PFTE-7糖酵解水平(60.30±3.46)mpH·min-1均高于癌旁细胞FT-6Tag(44.92±3.35)mpH·min-1、癌旁细胞FT-7Tag(44.06±2.76)mpH·min-1(P<0.05),卵巢癌细胞株A2780耗氧量(259.42±14.75)pmol·min-1、循环肿瘤细胞PFTE-6耗氧量(209.01±4.20)pmol·min-1和循环肿瘤细胞PFTE-7耗氧量(248.03±9.46)pmol·min-1高于癌旁细胞FT-6Tag(172.43±13.14)pmol·min-1、癌旁细胞FT-7Tag(124.29±7.82)pmol·min-1(P<0.05)。结论:循环肿瘤细胞的耗氧量及糖酵解较癌旁细胞强。
Objective:To observe the oxygen consumption and glycolysis of tumor cells in peripheral blood circulation of ovarian cancer patients.Methods:A total of 87 patients with ovarian cancer were collected.Density gradient centrifugation combined with immunomagnetic bead enrichment was used to screen circulating tumor cells in peripheral blood of ovarian cancer patients,and Seahorse was used to detect oxygen consumption and glycolytic capacity of circulating tumor cells.Results:Ovarian cancer cell line A2780(56.96±1.55),circulating tumor cells PFTE-6(61.67±5.13)and PFTE-7(60.30±3.46)had higher glycolytic capacity,and paracancerous cells FT-6Tag(44.92±3.35)and paracancerous cells FT-7Tag(44.06±2.76)had lower glycolytic capacity(P<0.05).Ovarian cancer cell line A2780(259.42±14.75),circulating tumor cells PFTE-6(209.01±4.20)and PFTE-7(248.03±9.46)were all higher in oxygen consumption than the adjacent cells FT-6Tag(172.43±13.14)and FT-7Tag(124.29±7.82)(P<0.05).To sum up,the oxygen consumption and glycolysis ability of circulating tumor cells were stronger than those of adjacent cancer cells.Conclusion:Oxygen consumption and glycolysis of circulating cancer cells are stronger than those of adjacent cancer cells,which may be a foundation for studying the invasion and metastasis of circulating tumor cells.
