微波辅助提取峨眉雪胆中雪胆甲素及其抑菌作用

Extraction of Cucurbitacin Ⅱa From Hemsleya emeiensis L. T. Shen by Microwave Assisted and Its Bacteriostasis

摘要 以峨眉雪胆为试材,采用单因素试验和正交实验对雪胆甲素的提取工艺进行优化,同时用滤纸片法测定雪胆甲素的抑菌活性,以期为研究峨眉雪胆中雪胆甲素的提取工艺及雪胆甲素的抑菌活性奠定基础。结果表明:峨眉雪胆中雪胆甲素的最佳提取条件为微波提取功率400 W、微波最佳提取时间为7 min、料液比1∶25 g·mL^-1、乙醇体积分数90%(v/v),此时提取率为3.8026 mg·g^-1。峨眉雪胆石油醚浸提液、乙酸乙酯浸提液和正丁醇浸提液在测定浓度范围内对大肠杆菌和金黄色葡萄球均有较好的抑菌活性。 Taking Hemsleya emeiensis L.T.Shen as raw material,single factor experiment and orthogonal experiment were used to optimize the extraction process of the Cucurbitacin Ⅱa,and the bacteriostatic of the Cucurbitacin Ⅱa was determined by filter paper.In order to lay foundation for the extraction technology and bacteriostasis of Cucurbitacin Ⅱa from Hemsleya emeiensis L.T.Shen.The results showed that the optimal extraction conditions were microwave optimal extraction power 400 W,microwave optimal extraction time 7 minutes,solid-liquid ratio 1∶25 g·mL^-1,ethanol concentration 90%(v/v),and the extraction rate was 3.802 6 mg·g^-1.The crude extracts of petroleum ether phase,ethyl acetate and n-butanol from Hemsleya emeiensis L.T.Shen showed good bacteriostasis to Escherichia coli and Staphylococcus aureus in the concentration range.
