摘要
Bex基因家族广泛分布于哺乳动物基因中,在细胞发育过程中有重要作用.以小鼠Bex基因家族5个成员(Bex1、Bex2、Bex3、Bex4、Bex6)为研究对象,通过生物信息学的研究方法,对该基因家族的基因结构、保守结构域、蛋白质性质进行系统分析,且绘制系统进化树.研究表明,染色体定位显示小鼠Bex基因主要分布在X染色体上(Bex1、Bex2、Bex3、Bex4),Bex6基因位于16号染色体;该家族所有的基因都有3个外显子及一个CDS区;小鼠Bex6基因可能是小鼠Bex4基因转座至16号染色体;有6个保守区域,具有motif 1、motif 2、motif 3、motif 4、motif 5、motif 6的定位分别为5、5、5、4、2、2个.6个motif的序列长度存在差异,分别为19、28、23、17、21、8.所有Bex蛋白不稳定,易降解,为亲水性蛋白,脂肪族指数在51.02~61.78之间,没有跨膜结构.Bex基因家族蛋白质等电点均不同,可用等点聚焦电泳区分.Bex蛋白亚细胞定位中,Bex1、Bex2位于细胞质和细胞核,Bex3、Bex4、Bex6位于细胞核.本研究为深入研究Bex基因家族提供理论依据.
Bex gene family is widely distributed in mammalian genes and plays an important role in cell development.On this study,5 members of musculus Bex gene family(Bex1,Bex2,Bex3,Bex4,Bex6)were taken as research objects.Through the research method of bioinformatics,the genetic structure,conserved domain and protein properties of the gene family were systematically analyzed,and the phylogenetic tree was drawn.Studies showed that the Bex gene was mainly distributed on the X chromosome(Bex1,Bex2,Bex3,Bex4),and Bex6 gene was located on chromosome 16.All the genes in the family have three exons and a CDS region.Musculus Bex6 gene may be the musculus Bex4 gene transposing to chromosome 16.There were six conservative regions with motif 1,motif 2,motif 3,motif 4,motif 5,motif 6 locations of 5,5,5,4,2 and 2,respectively.The six motifs have different sequence lengths,19,28,23,17,21,and 8,respectively.All Bex proteins are unstable,easily degraded and hydrophilic,with aliphatic index between 51.02 and 61.78 and no transmembrane structure.The isoelectric points of Bex gene family proteins are different,which can be distinguished by isotopic focusing electrophoresis.In subcellular localization,Bex1 and Bex2 are located in cytoplasm and nucleus,while Bex3,Bex4 and Bex6 are located in nucleus.This study provides theoretical basis for in-depth study of Bex gene family.
