八正散对急性肾盂肾炎模型大鼠炎症因子和局部免疫功能的影响 被引量:9

Effects of Bazheng Powder on Inflammatory Factors and Local Immune Function in Rats with Acute Py⁃elonephritis

摘要 目的探讨八正散对急性肾盂肾炎大鼠的治疗作用及可能机制。方法采用单侧输尿管结扎联合膀胱注射大肠杆菌建立急性肾盂肾炎大鼠模型,将36只造模成功大鼠随机分为模型组、八正散组和抗生素组,每组各12只,另取15只正常大鼠仅开腹不注射大肠杆菌为假手术组。造模3 d后,八正散组和抗生素组分别给予八正散和左氧氟沙星片灌胃,假手术组和模型组予以等体积的生理盐水灌胃。灌胃14 d后,计算肾脏质量比、肾脏指数、膀胱指数,检测尿和肾脏培养,尿白细胞计数、尿隐血,尿分泌性免疫球蛋白A(SIgA),尿肌酐,血清肌酐、白细胞计数(WBC)、降钙素原(PCT)、超敏C反应蛋白(hs-CRP)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白介素-10(IL-10)、趋化因子-2(CXCL-2)含量。结果与假手术组比较,模型组大鼠尿培养阳性率、肾脏培养阳性率、肾脏质量比、肾脏指数、膀胱指数、尿白细胞计数、尿隐血、尿肌酐、血肌酐、WBC、PCT、hs-CRP、IL-1β、IL-6、CXCL-2水平显著升高(P<0.05),IL-10、SIgA水平显著降低(P<0.05);与模型组比较,八正散组大鼠尿培养阳性率、肾脏培养阳性率、肾脏质量比、肾脏指数、膀胱指数、尿白细胞计数、尿隐血、尿肌酐、血肌酐、WBC、PCT、hs-CRP、IL-1β、IL-6、CXCL-2水平显著降低(P<0.05),IL-10、SIgA水平显著升高(P<0.05)。结论八正散可以抑制急性肾盂肾炎大鼠的炎症反应,提高局部免疫功能,抑制细菌生长。 Objective:To explore the therapeutic effect and possible mechanism of Bazheng Powder on acute py⁃elonephritis in rats.Methods:Unilateral ureteral ligation combined with bladder injection of Escherichia coli was used to establish acute pyelonephritis rat model;36 successfully modeled rats were randomly divided into model group,Bazheng Powder group and antibiotic group,12 rats in each group;another 15 normal rats were taken as the sham operation group with only opening abdominal surgery without injection of Escherichia coli.After 3 days of modeling,the Bazheng Powder group and the antibiotic group were given Bazheng Powder and levofloxacin tablets respectively,and the sham operation group and the model group were given an equal volume of normal saline.Af⁃ter gavage for 14 days,the kidney weight ratio,kidney index,bladder index were calculated;the urine and kidney culture,urine white blood cell count,urine occult blood(BLD),urine secretory immunoglobulin A(SIgA),urine cre⁃atinine,serum creatinine,white blood cells count(WBC),procalcitonin(PCT),high-sensitivity C-reactive protein(hs-CRP),interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),chemokine-2(CXCL-2)con⁃tent were checked.Results:In the model group,compared with the sham operation group,the positive rate of urine culture,positive rate of kidney culture,kidney weight ratio,kidney index,bladder index,urine white blood cell count,BLD,urine creatinine,blood creatinine,WBC,PCT,hs-CRP,IL-1β,IL-6,and CXCL-2 were signifi⁃cantly increased(P<0.05),and the levels of IL-10 and SIgA were significantly reduced(P<0.05);compared with the model group,the urine culture positive rate,kidney culture positive rate,kidney weight ratio,kidney index,bladder index,urine white blood cell count,BLD,urine creatinine,serum creatinine,WBC,PCT,the levels of hs-CRP,IL-1β,IL-6,CXCL-2 in the Bazheng Powder group were significantly reduced(P<0.05),and the levels of IL-10 and SIgA were significantly increased(P<0.05).Conclusion:Bazheng Powder can inhibit the inflammatory response in rats with acute pyelonephritis,improve local immune function,and inhibit bacterial growth.
