摘要
目的探讨枣仁安神胶囊对小鼠睡眠的作用及机制。方法昆明雄性小鼠,随机分为54组:空白对照组、枣仁安神胶囊低、中、高剂量组(0.19、0.38、0.76 g/kg),康斯顿阳性组(0.45 g/kg),连续30 d,灌胃给药。实验开始与结束,记录体重。采用协同戊巴比妥钠、巴比妥钠睡眠实验观察其对小鼠的催眠作用;小鼠脑组织中腺苷受体采用流式细胞仪和Western Blot检测。结果枣仁安神胶囊对小鼠体重增加没有影响,能延长戊巴比妥钠诱导小鼠睡眠时间,提高戊巴比妥钠阈下剂量小鼠入睡发生率,能减少巴比妥钠睡眠潜伏期。使小鼠脑组织中腺苷A1受体(A1 receptor,A1R)和A2A受体(A2A receptor,A2AR)含量增加。结论枣仁安神胶囊具有催眠作用,可能与增加A1R和A2AR的表达有关。
Objective To investigate the effect of Zaoren Anshen Capsule on the sleep in mice and its mechanism.Methods Male mice in Kunming were randomly divided into 54 groups:blank control group,Zaoren Anshen Capsule groups of low,medium and high dose(0.19,0.38,0.76 g/kg),and Conston positive group(0.45 g/kg).The mice received intragastric administration for 30 consecutive days.At the beginning and end of the experiment,their weight was recorded.The hypnotic effect on the mice was observed by the sleep experiment of synergetic pentobarbital sodium and barbiturate sodium.The adenosine receptor in brain tissue of the mice was detected by flow cytometry and Western Blot.Results Zaoren Anshen Capsule had no effect on the weight gain in the mice.It could prolong the sleep time of the mice induced by pentobarbital sodium,increase the incidence of sleep induced by subthreshold dose of pentobarbital sodium,and reduce the sleep latency of barbitol sodium,which led to the increase of adenosine A1 receptor(A1R)and A2A receptor(A2AR)in brain tissue of the mice.Conclusion Zaoren Anshen Capsule has hypnotic effect,which may be related to the increased expression of A1R and A2AR.
