摘要
目的探讨FK506对大鼠施万细胞株GFAP、NF200及Nogo-A蛋白表达的影响。方法依据实验需要大鼠RSC96施万细胞株分为:A组(空白对照组);B组(FK506诱导剂组);C组(DMSO溶剂对照组)。诱导1 d和14 d后用RT-PCR和免疫组织化学方法检测RSC96细胞株GFAP、NF200及Nogo-A的mRNA和蛋白的表达情况。结果在FK506处理14 d后,RSC96细胞GFAP、NF200的mRNA表达水平显著高于两对照组明显升高(P<0.01),而Nogo-A基因mRNA表达明显低于两对照组(P<0.01)。免疫组化检测显示,RSC96细胞株应用FK506处理14 d后,GFAP、NF200的阳性细胞数量明显多于两对照组(P<0.05),Nogo-A蛋白的阳性细胞数量,在诱导1 d时明显多于两对照组(P<0.05);而诱导14 d后,Nogo-A蛋白阳性细胞数量明显低于试剂对照组(P<0.05)。结论FK506可能具有促进GFAP和NF200表达的作用,同时可能具有抑制Nogo-A表达的作用。FK506可能具有促进大鼠施万细胞增殖分化的作用,从而加速周围神经损伤的修复与再生。
Objective To investigate the effect of FK506 on the expression of GFAP,NF200 and Nogo-A protein in Schwann cells of rats.Methods According to experimental requirements,rat RSC96wan cell lines were divided into:Group A(blank control group),Group B(FK506 inducer group)and Group C(DMSO solvent control group).MRNA and protein expressions of GFAP and NF200 and NOGO-A in RSC96 cell lines were detected by RT-PCR and immunohistochemistry 1 and 14 days after induction.Results After 14 days of FK506 treatment,the mRNA expression level of GFAP and NF200 in RSC96 cells was significantly higher than those in both control groups(P<0.01),and the mRNA expression of NOGO-A gene was significantly lower than those in both control groups(P<0.01).Immunohistochemical tests showed that after RSC96 cell line was treated with FK506 for 14 days,the number of positive cells of GFAP and NF200 was significantly higher than those of the two control groups(P<0.05);the number of Nogo-a protein positive cells was significantly higher than those of the two control groups 1 day after induction(P<0.05).After 14 days of induction,the number of Nogo-A protein-positive cells was significantly lower than that of Group C(P<0.05).Conclusion FK506 may promote the expression of GFAP and NF200,and inhibit the expression of Nogo-A.FK506 may promote the proliferation and differentiation of Schwann cells of rats,thus accelerating the repair and regeneration of peripheral nerve injury.
