反相高效液相色谱法测定龙葵果中澳洲茄碱和澳洲茄边碱的含量

Simultaneous Determination of Two Steroidal Alkaloids from the Fruit of Solanum Nigrum by RP-HPLC

利用反相高效液相色谱法快速测定龙葵果中2种甾体生物碱的含量。采用Agilent Extend-C 18(4.6 mm×150 mm,5μm)色谱柱,以乙腈-0.5%磷酸为流动相,流速1.0 mL/min,进样量5μL,柱温30℃,在203 nm条件下检测,以等度洗脱方式在30 min内分离了澳洲茄碱、澳洲茄边碱2种生物碱。结果表明,澳洲茄碱含量测定的线性范围为1.7~8.5μg(r=0.9996);澳洲茄边碱含量测定的线性范围为1.84~9.19μg(r=0.9999),平均回收率分别为100.18%和99.08%。该方法简便快速、准确度高,可用于龙葵药材的质量评价。

A new method for simultaneous determination of solasonine and solamargine in the fruit of Solanum Nigrum by reversed-phase HPLC was developed.The samples were separated at 30℃on Agilent Extend-C 18(4.6 mm×150 mm,5μm)column with acetonitrile-0.5%phosphoric acid as mobile phase.Flow rate was 1.0 mL/min and the detection wavelength was 203 nm.There was good linearity between the peak area and concentration at the ranges of 1.7~8.5μg(r=0.9996),1.84~9.19μg(r=0.9999)for solasonine and solamargine,respectively.The average recoveries of solasonine and solamargine were 100.18%and 99.08%.The method is rapid,simple and accurate,and it can be used for the evaluation of Solanum Nigrum L.
