胡椒酸乙二胺对高脂喂养大鼠血脂及肝形态结构的影响 被引量:2

Effect of piperic acid ethylenediamine on serum lipid profile and liver histology in rats fed with high fat diet

摘要 目的研究胡椒酸乙二胺(PAE)对高脂喂养大鼠肝脂肪变性及肝形态结构的影响。方法雄性SD大鼠65只,10只作为正常对照组基础饲料喂养,余高脂饲料喂养作为高脂模型,4周后将造模成功的50只高脂血症大鼠,随机分为高脂模型组,阳性药物组(10 mg/kg·bw),PAE高(10 mg/kg·bw)、中(5 mg/kg·bw)和低(2.5 mg/kg·bw)3个剂量组,每天灌胃给药,正常对照组和高脂模型组只给予0.5%羧甲基纤维素钠(CMC-Na)。干预4周后,处理动物,采集血清分析比较各组大鼠血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)和低密度脂蛋白(LDL-C)水平;采集肝组织,光学显微镜观察肝形态结构。结果与正常对照组和高脂模型组相比,PAE各剂量组均能降低TC、TG和LDL-C水平,升高HDL-C水平,差异有统计学意义(P<0.05);与阳性药物组比较,PAE高剂量组LDL-C降低水平差异有统计学意义(P<0.05)。肝病理形态学结果显示,PAE各干预组明显改善由高脂饮食诱导的肝细胞形态结构改变和脂肪浸润,以PAE高剂量组优于模型组和药物对照组。结论 PAE可调节高脂喂养大鼠血脂异常,并减少肝损伤,其药效学作用还有待进一步研究。 Objective To investigate the effect of piperic acid ethylenediamine(PAE) on hepatic steatosis in rats fed with high-fat diet. Methods Sixty-five male Sprague-Dawley rats, 10 were fed with basic normal feed as the normal control group, and the rest were fed with high-fat diet. After four weeks of feeding, the rat model with hyperlipidemia rats were randomly divided into high-fat group, positive drug group(10 mg/kg·bw), and high(10 mg/kg·bw), medium(5 mg/kg·bw), low(2.5 mg/kg·bw) dose of PAE groups. The rats were intragastrically administered with drug or different dose of PAE daily, and the normal control group and the high fat group were given the same volume of 0.5% CMC-Na solution. Four weeks after intervention, all animals were sacrificed, and serum samples were collected to compare total cholesterol(TC), triglyceride(TG), high-density lipoprotein(HDL-C), and low-density lipoprotein(LDL-C) levels in different group. Liver tissue was collected to observe structure by light microscope. Results Compared with the normal control group and the high-fat model group, the levels of TC, TG, LDL-C were decreased, and the HDL-C level was increased in all PAE dose group and the difference was statistically significant(P<0.05);Compared with the positive drug group, the LDL-C level in the high dose group of PAE was significantly lower(P<0.05). Pathological changes in the liver tissue showed that different doses of PAE significantly improved the morphological structure and fat infiltration in liver of rats induced by high-fat feeding, and the PAE high dose group was superior to the high-fat model group and the drug control group. Conclusion PAE can regulate lipid metabolism and decrease liver damage in high-fat feeding rats.
