摘要
目的观察葡萄籽原花青素(grape seed proanthocyanidin extract,GSPE)对D-半乳糖致衰老模型小鼠的抗衰老作用。方法将小鼠随机分为空白组、模型组、GSPE低、中、高剂量组、维生素C阳性对照组,采取注射D-半乳糖30 d致小鼠衰老模型,灌胃给药治疗60 d后,测定小鼠脾脏、胸腺、肝脏指数;生化法测定小鼠肝组织、血液中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)含量及皮肤组织中SOD、GSH-Px、羟脯氨酸(HYP)含量;HE染色观察肝脏形态变化。结果 GSPE中、高剂量组脾脏、胸腺、肝脏指数显著改善,肝组织、血液及皮肤组织中SOD、GSH-Px含量显著增加,MDA及HYP含量显著降低。肝细胞形态和排列趋于正常,炎症情况减轻。结论 GSPE对D-半乳糖致衰老模型小鼠氧化应激反应的各项生化指标及肝脏组织病理状态有一定的改善作用。
Objective To observe the anti-aging effect of grape seed proanthocyanidin extract(GSPE) on D-galactose induced aging mice. Methods The mice were divided into blank group, model group, low-, medium-and highdose of GSPE groups and vitamin C positive control group. The aging model of mice was induced by injection of D-galactose for 30 days, and the mice were treated by gavage for 60 days. Then the spleen, thymus and liver indexes were measured;the levels of SOD, GSH-PX and MDA in liver tissues and blood, and the levels of SOD, GSH-Px and HYP in skin tissues were measured by biochemical method;and the morphological changes of the liver tissues were observed by HE staining. Results The spleen, thymus and liver indexes were significantly improved by treatment with GSPE;the levels of SOD and GSH-Px in liver tissues, blood and skin tissues increased, while the levels of MDA and Hyp significantly decreased. Besides, The morphology and arrangement of hepatocytes tended to be normal, and the inflammation was alleviated. Conclusion GSPE can improve the biochemical indexes and hepatic histopathological state of aging mice induced by D-galactose.
