HPLC同时测定杜仲平压胶囊中4种成分的含量

摘要 目的建立同时测定杜仲平压胶囊中桃叶珊瑚苷、绿原酸、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷4个指标性成分含量的高效液相色谱(HPLC)法。方法采用HPLC测定杜仲平压胶囊中桃叶珊瑚苷、绿原酸、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷含量,色谱柱:Diamonsil?PlusC18(250 mm×4.6 mm,5μm);流动相:甲醇(A)-0.05%磷酸水溶液(B),梯度洗脱;流速:1 ml/min;柱温:25℃;进样量:10μl;程序变化波长:10~15 min:210 nm(检测桃叶珊瑚苷),16~20 min:320 nm(检测绿原酸),30~35 min:230 nm(检测松脂醇二葡萄糖苷),35~40 min:230 nm(检测松脂醇单葡萄糖苷)。结果方法的专属性、精密度、重复性、稳定性均符合要求,线性范围分别为0.0123~1.235 mg/ml(R2=0.9999),0.0312~3.1250 mg/ml(R2=0.9999),0.0067~0.0976 mg/ml(R2=0.9999),0.0052~0.5231 mg/ml(R2=0.9998);加样回收率分别为100.66%(RSD=1.22%),100.87%(RSD=0.60%),101.33%(RSD=2.22%),100.88%(RSD=1.90%)。结论本方法简便、稳定、快速、重现性好,可同时测定杜仲平压胶囊中桃叶珊瑚苷、绿原酸、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷4个指标成分含量,为杜仲平压胶囊质量控制提供一定参考。 Objective To establish a high performance liquid chromatography(HPLC)method for the simultaneous determination of aucubin,chlorogenic acid,pinoresinol diglucoside and terpineol monoglucoside in Duzhong Pingya Capsules.Methods The contents of aucubin,chlorogenic acid,pinoresinol diglucoside and terpineol monoglucoside were determined by HPLC.The separation was performed on Diamonsil®Plus C18 column(250 mm×4.6 mm,5μm),using methanol(A)-0.05%phosphoric acid aqueous solution(B)as mobile phase of gradient elution.The flow rate was 1 ml/min;the column temperature was 25℃;the injection volume was 10μm.The detection wavelength was programmed as follows:10-15 min(210 nm,for determination of aucubin),16-20 min(320 nm,for determination of chlorogenic acid),30-35 min(230 nm,for determination of pinoresinol diglucoside),35-40 min(230 nm,for determination of terpineol monoglucoside).Results The specificity of the method met the relevant requirements.The linear ranges of aucubin,chlorogenic acid,pinoresinol diglucoside and terpineol monoglucoside were 0.01231.235 mg/ml(R^2=0.9999),0.0312-3.1250 mg/ml(R^2=0.9999),0.0067-0.0976 mg/ml,(R^2=0.9999)and 0.0052-0.5231 mg/ml(R^2=0.9998),respectively.The average recoveries were 100.66%(RSD=1.22%),100.87%(RSD=0.60%),101.33%(RSD=2.22%)and 100.88%(RSD=1.90%),respectively.Conclusion The method is simple,stable,rapid and reproducible.It can be used for determination of aucubin,chlorogenic acid,pinoresinol diglucoside and terpineol monoglucoside in Duzhong Pingya Capsules at the same time,and can provide a reference for the quality control of Duzhong Pingya Capsules.
