摘要
目的基于六味补血胶囊16组拆方的全息化学信息和对血虚小鼠造血功能的影响,分析其谱效相关性,为明确六味补血胶囊的药效物质基础提供实验依据。方法采用HPLC法测定六味补血胶囊16组拆方的全息化学信息,并对色谱峰进行药味归属;建立与中医临床接近的血虚动物模型,研究不同组方对环磷酰胺所致血虚小鼠外周血象指标(血红蛋白、红细胞、网织红细胞)的影响;用灰色关联度法分析两者的相关性。结果关联度分析结果显示,六味补血胶囊造血功能的药效作用是多种成分共同作用的结果,关联度均大于0.550。峰2(5-羟甲基糠醛,熟地黄)、峰11(当归、川芎)、峰12(藁本内酯,当归、川芎)对3个药效指标的贡献均排在前3位,关联度均大于0.7,是造血功能的主要有效成分。结论通过化学成分与药效结果的相关性研究,明确了六味补血胶囊的药效物质基础,为选择与药效相关的代表性成分并建立科学合理的质控标准提供参考依据。
Objective To analyze the spectrum-effect relationship based on the holographic chemical information and the effect on the hematopoietic function of blood deficiency mice of the 16 demolition groups of Liuwei Buxue Capsule,and to provide experimental basis for the determination of the pharmacodynamic material basis of Liuwei Buxue Capsule.Methods The holographic chemical information of the 16 demolition groups of Liuwei Buxue Capsule was determined by HPLC,and the chromatographic peaks were identified and matched with Chinese Materia Medica in the prescription.An animal model of blood deficiency was established to study the effects of different formulations on peripheral blood parameters(hemoglobin,red blood cell and reticulocytes) of mice with blood deficiency caused by cyclophosphamide.The correlation between them was analyzed by grey relational degree method.Results Correlation analysis results showed that the therapeutic effects of Liuwei Buxue Capsule on hematopoietic function were a result of a variety of ingredients,correlation degrees were greater than 0.550.No.2 peak(5-hydroxymethylfurfural,Rehmannia Glutinosa),No.11 peak(Angelicae sinensis radix and Chuanxiong Rhizoma),No.12 peak(ligustilide,Angelicae sinensis radix and Chuanxiong Rhizoma) were the top three compounds contributing to the three efficacy indices,correlation degrees were greater than 0.7,so they were the main active ingredients of hematopoietic function.Conclusion Based on the correlation study of chemical compounds and pharmacodynamic results,the pharmacodynamic material basis of Liuwei Buxue Capsule is clarified,and to provide reference basis for selecting representative components related to pharmacodynamics and establishing scientific and reasonable quality control standard.
