小凹蛋白3和HCN4在心力衰竭兔窦房结的表达变化

摘要 目的 探讨兔心力衰竭后心功能和小凹蛋白3(caveolin-3)及HCN4的蛋白表达的变化.方法 采用主动脉瓣反流联合腹主动脉缩窄的方法制作慢性心力衰竭模型.通过透射电镜观察心力衰竭前后兔窦房结组织小凹的变化,通过western plot研究心力衰竭后兔窦房结caveolin-3和HCN4的蛋白表达变化.结果 与对照组相比,心力衰竭组LVESD、LVEDD、LVESV和LVEDV值较对照组均升高[LVESD(14.33±2.64)mm比(7.95±0.42)mm;LVEDD(19.67±3.59)mm比(12.68±1.09)mm;LVESV(2.71±0.97)ml比(1.24±0.35)ml;LVEDV(8.42±2.03)ml比(5.73±0.89)ml](P均<0.05),而LVEF和LVFS值降低[LVEF(41.23±5.38)%比(78.37±8.13)%,LVFS(21.29±1.76)%比(43.48±5.22)%,P<0.05].心力衰竭后窦房结小凹丰度下降.心力衰竭显著降低窦房结caveolin-3和HCN4的蛋白表达水平,且caveolin-3表达下降与HCN4表达降低有相关性(r=0.83,P<0.05).结论 心力衰竭后窦房结小凹数量减少导致定位在小凹上HCN4通道表达改变,窦房结HCN4表达下调可能与caveolin-3的降低相关.这些发现为心脏冲动形成的正常和疾病相关损伤的分子基础和调控提供了新的依据. Objective To investigate the change of function and the protin expressions of caveoli and HCN4 in rabbit heart failure(HF).Methods HF was produced by combined aortic isufficicy and abdominal aortic constiction.The change of caveolae in sinus node tssue was observed by transmission eleton microscopy.Westem plot was used to observe the change of the protein expression of caveoli and HCN4.Resuts Compared with the control group,the LVESD,LVEDD,LVESV and LVEDV values were all increased in heart fiure group [LVESD(14.33±2.64)mm vs (7.95±0.42)mm;LVEDD(19.67±3.59)mm vs(12.68±1.09)mm;LVESV(2.71±0.97)vs.(1.24±0.35)ml;LVEDV(8.42±2.03)ml vs.(5.73±0.89)m](P<0.05),while the LVEF and LVFS values lower[LVEF(41.23±5.38)%vs.(78.37±8.13)%,LVFS(21.29±1.76)%vs.(43.4±85.22)%,P<0.05].Protein expression levels of caveolin-3 and HCN4 significantly reduced affer heart failure and HCN4 abundancce psitively crelted wih the rductio of cvoeli-3(r=0.83,P<0.0).Concusion Reduction of cavcolac number leads to the xressson change of HCN4 chane which location in cavole dow-reguation of HCN4 expessso might cotriute to CHF induced sius node dyfuction These finding provide novel information about the molecular basis of normal and disease related impaiments of cardiac impulse formation.
