流苏石斛多糖抗氧化活性研究 被引量:5

摘要 为了研究流苏石斛多糖(polysaccharides from Dendrobium fimbriatum Hook.,DFP)的体内外抗氧化活性作用,文章提取流苏石斛茎多糖,测定流苏石斛多糖对ABTS、DPPH和·OH自由基的清除能力;采用四氧嘧啶腹腔注射法建立糖尿病小鼠模型,测定流苏石斛多糖对小鼠肾脏和肝脏超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活力及还原型谷胱甘肽(GSH)和丙二醛(MDA)量的影响。流苏石斛多糖具有清除ABTS、DPPH和·OH自由基的能力,其体外抗氧化活性与多糖质量浓度呈现良好的量效关系;流苏石斛多糖能显著提升肝脏中SOD和GSH水平以及肾脏中CAT和GSH水平,有效降低糖尿病小鼠肝脏和肾脏中MDA的量;流苏石斛多糖能够在体内和体外发挥抗氧化作用,但在不同器官中发挥抗氧化活性的机制不同。 The antioxidative activities in vivo and in vitro of polysaccharides from Dendrobium fimbriatum Hook.(DFP)were investigated.DFP was extracted from the stem of D.fimbriatum.The scavenging ability of DFP on ABTS,DPPH and hydroxyl free radicals was determined.Diabetic mouse model was established by intraperitoneal injection of alloxan.The enzyme activities of SOD and CAT as well as the contents of GSH and MDA of liver and kidney in mice were determined.DFP had the ability to scavenge ABTS,DPPH and hydroxyl free radicals,and showed a good dose-dependent effect with the polysaccharide concentrations.DFP could effectively decrease MDA content and enhance the levels of SOD and GSH in hepatic tissues of diabetic mice.DFP could also depress the increase in MDA content and raise the levels of CAT and GSH in renal tissues.DFP possessed antioxidative activities in vivo and in vitro,and its antioxidative function was different in different tissues.
