摘要
目的探讨盐炙杜仲对维甲酸诱导的骨质疏松大鼠骨代谢的影响及作用机制。方法将60只3月龄♀SD大鼠按随机数字表法分为六组:空白组、模型组、阳性对照组,杜仲低、中、高剂量组(DL/DM/DH)。除空白组外,其余各组灌胃维甲酸,建立骨质疏松模型,空白组灌胃同等剂量的0.5%CMC-Na溶液,持续2周。造模成功后,各给药组大鼠分别灌胃相应药物,模型组和空白组灌胃等体积0.5%CMC-Na溶液,每天1次,持续灌胃4周后,检测六组大鼠血清1型胶原N前端肽(PINP)、1型胶原C末端肽(CTX)、骨保护素(OPG)、雌二醇(E_2)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)、骨密度(BMD)的变化。结果与空白组相比,模型组大鼠BMD、OPG及E_2水平明显下降(P <0.05),TNF-α、IL-6、PINP及CTX水平显著升高(P <0.05)。给药后,阳性对照组及杜仲中、高剂量组大鼠BMD、OPG及E_2水平有显著改善(P <0.05),TNF-α、IL-6、PINP及CTX水平有明显降低趋势。结论中、高剂量盐炙杜仲可以通过调节骨转换状态,影响激素分泌及细胞因子的表达,达到防治骨质疏松的作用。
Objective To observe the effects of salt-processed eucommia on bone metabolism in retinoic acid-induced osteoporosis rats and its mechanism.Methods 60 female SD rats were randomly divided into six groups:blank control group,model group,positive control group,eucommia low/medium/high dose group(DL/DM/DH).Except the blank group,the other groups were given retinoic acid to establish the osteoporosis model,and the blank group was given 0.5%CMC-Na solution with the same dose for 2 weeks.After successful modeling,each dose group rats respectively corresponding drug lavage,model group and blank group and gastric volume 0.5%CMC-Na solution,once a day.After 4 weeks,detection of six groups of rats PINP/CTX/OPG/E2/IL-6/TNF and the change of BMD.Results Compared with the blank group,the BMD,OPG and E2 levels of the model group were significantly decreased(P<0.05),and the levels of TNF-α,IL-6,PINP and CTX were significantly increased(P<0.05).After administration,the BMD,OPG and E2 levels of rats in the positive control group and the middle and high dose groups of Eucommia ulmoides significantly improved(P<0.05),and the levels of TNF-α,IL-6,PINP and CTX showed a significant decrease.Conclusion The experiment shows that middle and high doses of eucommia ulmoides can regulate the bone turnover state,affect hormone secretion and cytokine expression,and achieve the effect of preventing osteoporosis.
