鸡CPT1A基因的生物信息学分析和组织表达谱构建 被引量:1

Bioinformatics of chicken CPT1A gene and construction of its expression profile in tissue

摘要 旨在探索鸡肉碱棕榈酰转移酶1A(CPT1A)基因的生物学特征,研究其表达的组织和时空差异。通过生物信息学软件对CPT1A序列进行分析和功能预测,利用荧光定量PCR技术检测该基因在12周固始鸡腹脂、皮脂、胸肌、腿肌和肝中的表达量,以及在不同时期(6周,12周,22周和30周)固始鸡胸肌中的表达量。结果显示,鸡CPT1A蛋白无信号肽,有两个跨膜结构域。鸡与斑马鱼CPT1A基因的亲缘关系最远,与小鼠、人、猪、山羊、奶牛的亲缘关系较近。CPT1A基因在12周固始鸡肝脏组织中表达量最高,胸肌和腿肌次之,腹脂和皮脂中表达量较低,在不同发育阶段中固始鸡胸肌中表达量相对较高的为12周和6周,22周次之,30周则表达量最低。试验结果为进一步研究鸡CPT1A基因调控鸡脂质代谢的分子机制提供理论基础。 In order to explore the biological characteristics of the chicken base palmitoyltransferase 1 A(CPT1 A)gene,and its expression in the tissue and temporal and spatial differences in the expression,we analyzed its sequence and performed function prediction through bioinformatics software,and used fluorescent quantitative PCR to detect the expression levels of the gene in the abdominal fat,sebum,breast muscle,leg muscles and liver in Gushi chickens at different stages of growth(6 weeks,12 weeks,22 weeks and 30 weeks).The results showed that the chicken CPT1 A protein had no signal peptide but showed two transmembrane domains.The CPT1 A gene of the chickens was of the farthest genetic relationship with that zebrafish,but was quite closely related to that of mice,humans,pigs,goats,and dairy cows.The expression of the gene was the highest in liver tissue of the Gushi chickens at 12 weeks of age,followed by the expression in their breast,leg muscles,abdominal fat and sebum.Of the four stages of growth,the highest expression of the gene in the breast muscle of the Gushi chickens was at 12 weeks,followed by 6 weeks and 22 weeks,and the lowest at 30 weeks of age.The present results provided a theoretical basis for further research on the molecular mechanism of chicken CPT1 A gene in regulating lipid metabolism in chickens.
