摘要
目的探讨头穴丛刺对VaD大鼠学习记忆能力及Bcl-2、Bax、Caspase-3表达的影响。方法将SD大鼠随机分为空白组、假手术组、模型组和头穴丛刺组,双血管阻断法制备大鼠VaD模型。头穴丛刺组予头穴丛刺针刺干预,连续干预28 d。Morris水迷宫实验测试大鼠的学习和记忆能力。免疫组化染色法检测bax、Bcl-2、Caspase-3表达量。结果与假手术组比较,模型组逃避潜伏期显著延长(P<0.01);与模型组比较,头穴丛刺组逃避潜伏期明显缩短(P<0.01)。与假手术组比较,模型组目标象限停留时间显著缩短(P<0.01),跨越平台次数明显降低(P<0.01);与模型组比较,头穴丛刺组目标象限停留时间明显延长(P<0.01)、跨越平台次数增加(P<0.01)。与假手术组比较,模型组Bcl-2表达弱(P<0.01),Bax、Caspase-3表达显著增加(P<0.01);与模型组比较,头穴丛刺组Bcl-2表达增加(P<0.01),Bax、Caspase-3表达减弱(P<0.01)。结论头穴丛刺可改善VaD大鼠学习和记忆能力,能够上调Bcl-2表达、下调Bax、Caspase-3表达。
Objective To study the effects of scalp cluster thorn on learning and memory ability and expression of Bcl-2,Bax and Caspase-3 in VaD rats.Methods SD rats were randomly divided into blank group,sham operation group,model group and cluster acupuncture group.In the group of cluster acupuncture at the head points,the patients were given the intervention of cluster acupuncture at the head points,which lasted for 28 days.Morris water maze experiment tested the learning and memory ability of rats.The expression levels of Bax,Bcl-2 and Caspase-3 were detected by immunohistochemical staining.Results Compared with the sham operation group,the escape latency of the model group was significantly prolonged(P<0.01).Compared with the model group,the escape latency of rats in the cluster acupuncture group was significantly shortened(P<0.01).Compared with the sham operation group,the target quadrant residence time of the model group was significantly shortened(P<0.01),and the number of crossing the platform was significantly reduced(P<0.01).Compared with the model group,the target quadrant residence time of rats in the cluster acupuncture group at the first point was significantly prolonged(P<0.01),and the number of crossing the platform was increased(P<0.01).Compared with the sham group,Bcl-2 expression was weak in the model group(P<0.01),while Bax and Caspase-3 expression were significantly increased in the model group(P<0.01).Compared with the model group,Bcl-2 expression increased(P<0.01),and Bax and Caspase-3 expression decreased(P<0.01).Conclusion Scalp cluster thorn can improve the learning and memory ability of VaD rats,and up-regulate the expression of Bcl-2 and down-regulate the expression of Bax and Caspase-3.
