UPLC-MS-MS法同时测定玉屏风颗粒中6种皂苷类成分 被引量:5

Simultaneous determination of 6 saponins in Yupingfeng granules by UPLC-MS-MS

摘要 目的建立同时测定玉屏风颗粒中6种皂苷类成分的快速方法,以更全面地评价玉屏风颗粒的质量。方法采用UPLC-MS-MS法,ESI离子源,色谱柱为Waters ACQUITY UPLC HSS T3 C18(1.8μm,2.1 mm×100 mm),流动相为乙腈-0.1%甲酸(梯度洗脱),流速为0.30 mL/min,柱温为30℃,进样量为2μL。结果黄芪甲苷、黄芪皂苷Ⅲ、黄芪皂苷Ⅱ、异黄芪皂苷Ⅱ、黄芪皂苷Ⅰ和异黄芪皂苷Ⅰ质量浓度分别在0.1775~3.55、0.0625~1.25、0.298~5.96、0.1575~3.15、0.2025~4.05、0.29~5.8μg/mL范围内与峰面积呈良好的线性关系,平均回收率分别为100.64%、97.66%、100.89%、104.57%、101.73%、98.19%,RSD分别为3.54%、1.93%、2.63%、2.42%、3.35%、2.57%。结论本文方法同时对玉屏风颗粒的6个皂苷类成分进行分析,方法简便、快速、准确,为有效评价玉屏风颗粒质量提供实验依据。 Objective To establish a simultaneous determination method for 6 saponins in Yupingfeng granules,for the purpose of more precisely evaluation of its quality information.Methods UHPLC-MS-MS was adopted on a Waters ACQUITY UPLC HSS T3 C18(1.8μm,2.1 mm×100 mm)column,and acetonitrile-0.1%formic acid was used as the mobile phase by gradient elution.The flow rate was 0.3 mL/min,the column temperature was set at 30℃,and the injection volume was 2μL.Results Astragaloside,AstragalosideⅢ,AstragalosideⅡ,IsostragalosideⅡ,AstragalosideⅠand IsostragalosideⅠhad a good linearity relationship between 0.1775-3.55,0.0625-1.25,0.298-5.96,1575-3.15,0.2025-4.05,0.29-5.8μg/mL,and the average recoveries were 100.64%(RSD=3.54%),97.66%(RSD=1.93%),100.89%(RSD=2.63%),104.57%(RSD=2.42%),101.73(RSD=3.35%)and 98.19%(RSD=2.57%),respectively.Conclusion The method of simultaneous determination for 6 saponins in Yupingfeng granules is simple,rapid and accurate,which provides experimental basis for the quality evaluation of Yupingfeng granules.
