中药复方DC对标准大肠杆菌、产ESBLs大肠杆菌的体内外抗菌作用研究 被引量:2

摘要 目的探索中药复方DC对标准大肠杆菌、产ESBLs大肠杆菌的体内外抗菌作用,为研发替代抗生素中药抗耐药菌新药提供数据参考。方法体外抗菌以二倍稀释法、纸片法测定中药复方DC对标准大肠杆菌及产ESBLs大肠杆菌最低抑菌浓度、抑菌圈直径;体内抗菌通过观察中药复方DC低、中、高(4.5、9、18 g/70 kg)剂量给药5 d对小鼠的致死保护率,同时测定小鼠血清中IL-6、IFN-γ含量。结果中药复方DC对标准大肠杆菌、产ESBLs大肠杆菌的最低抑菌浓度分别为25.0、25.0 mg/mL,抑菌圈直径分别为23.0、24.0 mm。标准大肠杆菌感染:与模型组相比,中药复方DC高、中剂量组、氨苄西林组在小鼠死亡率、小鼠血清中IL-6和IFN-γ含量方面的差异均具有统计学意义(P<0.01或P<0.05)。产ESBLs大肠杆菌感染:与模型组相比,中药复方DC高剂量组、亚胺培南组在小鼠死亡率、小鼠血清中IL-6和IFN-γ含量方面的差异均具有统计学意义(P<0.01或P<0.05)。结论中药复方DC对标准大肠杆菌、产ESBLs大肠杆菌具有较好的体内外抑制作用,同时可对感染小鼠起到抗炎作用。 Objective To elucidate the antibacterial effect of Chinese herbal compound DC on standard E.coli and ESBLs producing E.coli in vitro and in vivo,and provide references for the development of alternative antibiotics of traditional Chinese medicine against drug-resistant bacteria.Methods The minimum inhibitory concentration(MIC)and diameter of inhibition zone of Chinese herbal compound DC against standard E.coli and ESBLs producing E.coli were determined by double dilution method and disk method.The lethal protective rates of Chinese herbal compound DC at low,medium and high doses(4.5,9,18 g/70 kg)for 5 days in mice were observed.The serum contents of IL-6 and IFN-γwere determined.Results The MIC of cmpound DC against standard E.coli and ESBLs producing E.coli were 25.0 mg/mL and 25.0 mg/mL,and the diameters of inhibition zone were 23.0 mm and 24.0 mm,respectively.Standard E.coli infection:Compared with the model group,there were significant differences in the mortality of mice and the contents of serum IL-6 and IFN-γin the high-dose group,the middle dose group and the ampicillin group(P<0.01 or P<0.05).ESBLs producing E.coli infection:Compared with the model group,there were significant differences in the mortality of mice and the serum contents of IL-6 and IFN-γin high-dose DC group and imipenem group(P<0.01 or P<0.05).Conclusion The Chinese herbal compound DC shows good inhibitory effect on standard E.coli and ESBLs producing E.coli in vitro and in vivo,and it also has anti-inflammatory effect on infected mice.
