依托咪酯通过调节Hippo信号通路抑制鼻咽癌细胞CNE-1和HNE-1的侵袭迁移 被引量:3

摘要 目的探究依托咪酯通过调节Hippo信号通路对鼻咽癌CNE-1和HNE-1细胞侵袭迁移的作用。方法MTT法检测细胞活性;对照组、依托咪酯低、中、高剂量组鼻咽癌CNE-1和HNE-1细胞用终浓度含有依托咪酯0、10、20、40μg/mL培养基培养;顺铂组鼻咽癌CNE-1和HNE-1细胞用终浓度含有Cisplatin 10μg/mL培养基培养;Hippo通路抑制剂组和依托咪酯高剂量+XMU-MP-1组鼻咽癌CNE-1和HNE-1细胞用终浓度含有XMU-MP-110μg/mL和依托咪酯0、40μg/mL培养基培养。划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭,蛋白质印迹分析检测p-MST、p-LATS、p-YAP、MST、LATS、YAP蛋白表达水平。结果选择依托咪酯10、20、40μg/mL和药物作用时间24 h作后续实验。高浓度的依托咪酯减少鼻咽癌CNE-1和HNE-1细胞迁移率和侵袭能力,上调p-MST/MST、p-LATS/LATS、p-YAP/YAP蛋白表达(P<0.01)。加入Hippo信号通路抑制剂逆转依托咪酯对鼻咽癌CNE-1和HNE-1细胞迁移和侵袭作用。结论依托咪酯通过调节Hippo信号通路抑制鼻咽癌CNE-1和HNE-1细胞的侵袭迁移。 Objective To investigate the effect of etomidate on the invasion and migration of nasopharyngeal carcinoma CNE-1 and HNE-1 cells by regulation of the Hippo signaling pathway.Methods Cell activity was detected by MTT method.CNE-1 and HNE-1 cells in the control,low,medium,and high dose groups of etomidate were cultured with etomidate at 0,10,20 and 40μg/mL.CNE-1 and HNE-1 cells in the cisplatin group were cultured with 10μg/mL Cisplatin.CNE-1 and HNE-1 cells in the Hippo pathway inhibitor group and the high-dose etomidate+XMU-MP-1 group were treated with 10μg/mL XMU-MP-1 and 40μg/mL etomidate medium.Cell migration ability was detected by scratch test,cell invasion was detected by Transwell test,and the expression of p-MST,p-LATS,p-YAP,MST,LATS and YAP protein were detected by western blot.Results Etomidate at the doses of 10,20,40μg/mL and drug action time 24 h were selected for subsequent experiments.Etomidate at a high concentration reduced the migration and invasion capacity of CNE-1 and HNE-1 cells,and up-regulated the expression of p-MST/MST,p-LATS/LATS and p-YAP/YAP protein(P<0.01).In addition,Hippo signaling inhibitor reversed the effects of etomidate on the migration and invasion of CNE-1 and HNE-1 cells.Conclusion Etomidate inhibits the invasion and migration of nasopharyngeal carcinoma CNE-1 and HNE-1 cells by regulating the Hippo signaling pathway.
