摘要
目的了解绍兴地区艾滋病病毒(HIV)基因亚型分布特点,建立本地区HIV-1分子生物学及流行病学数据库。方法回顾性分析2013年1月-2014年7月本地区新确认的100例感染者样本,采用RT-PCR和nest-PCR方法扩增HIV-1的gag/pol区全长基因并测序,基因序列构建Neighbor-Joining系统树分析确定基因亚型。结果共获得82例样品的序列结果,发现CRFO1_AE,CRFO7_BC、CRFO8_BC和B4种亚型,各占46.34%、,35.37%,9.76%和8.54%;经性传播感染HIV/AIDS达96.34%(79例),不同途径感染的HIV-1亚型差异无统计学意义。CRFO7_BC,CRFO8_BC亚型与云南及广西病毒株相近,B亚型则与河南输血传播病毒株相近,CRFO1_AE亚型分为两簇,一簇与福建株相近,另一簇与北京、辽宁MSM病毒株相近。结论―绍兴地区HIV主要通过性途径传播,主要流行的亚型为CRFO1_AE和CRF07_BC。
Objective To understand the distribution characteristics of HIV gene subtypes in Shaoxing,and to establish the molecular and epidemiological database of HIV-1 in Shaoxing. Methods Retrospective analysis was performed on 100 newly confirmed infected patients in the region from January 2013 to July 2014. The full-length gag/pol gene of HIV-1 was amplified by RT-PCR and nest-PCR,and then sequenced. Neighbor-Joining system tree was constructed to analyze and determine the gene subtypes. Results A total of 82 samples were sequentially obtained. CRF01_AE,CRF07_BC,CRF08_BC and B subtypes were found,accounting for 46. 34%,35. 37%,9. 76% and 8. 54% respectively. The sexually transmitted infection of HIV/AIDS reached 96. 34%( 79 cases),and there was no statistical significance on the difference in the subtype of HIV-1 infected by different routes. The CRF07_BC and CRF08_BC subtypes were similar to Yunnan and Guangxi virus strains,while the B subtypes were similar to Henan transfusion-transmitted virus strains. The CRF01 _AE subtypes were divided into two clusters,one cluster was similar to Fujian strain and the other cluster was similar to Beijing and Liaoning MSM virus strains.Conclusion HIV is mainly transmitted through sexual channels in Shaoxing,and the main epidemic subtypes are CRF01_AE and CRF07_BC.
