摘要
目的探讨蜂胶小分子凝胶对老年大鼠口腔溃疡的治疗作用及机制。方法50只雄性大鼠(12月龄),随机分为5组,每组10只,采用40%冰醋酸灼烧法制备老年大鼠口腔溃疡模型,模型组(A组):不给予药物治疗。膜剂组(B组):给予市售紫竹药业蜂胶口腔膜,每天1次,每次半贴。蜂胶微粉组(C组):给予蜂胶提取物微粉,每天1次,每次1 mg。小分子凝胶基质组(D组):给予不载蜂胶提取物微粉的小分子凝胶基质,每天1次,每次半支。蜂胶小分子凝胶组(E组):给予蜂胶小分子凝胶,每天1次,每次半支。各组大鼠采用10%水合氯醛麻醉后给药,给药前后用电子游标卡尺测量口腔溃疡直径,观察每组大鼠的体重变化,记录各组大鼠的愈合时间。通过酶联免疫吸附试验检测口腔溃疡老年大鼠在治疗第1天、第3天和第7天血清中白细胞介素(IL)-2、IL-6和肿瘤坏死因子(TNF)-α水平。结果治疗前、治疗后第1天和第2天各组口腔溃疡直径比较差异无统计学意义(P>0.05),治疗后第3天,B、C、E组口腔溃疡直径与A组比较差异有统计学意义(P<0.05),治疗后第4天B组、C组、D组、E组口腔溃疡直径与A组比较差异有统计学意义(P<0.01)。造模后各组大鼠体重均有明显下降趋势,A组第7天开始回升,B组、C组第4天开始回升,D组第6天开始回升,E组第3天体重开始回升。B组、C组、D组、E组口腔溃疡平均愈合时间明显小于A组(P<0.05,P<0.01),B组、C组和E组口腔溃疡平均愈合时间与D组比较差异有统计学意义(P<0.05,P<0.01),B组、C组和E组口腔溃疡平均愈合时间比较差异无统计学意义(P>0.05)。随给药时间的延长,A组、B组和E组TNF-α和IL-6的水平逐渐减低,IL-2的水平逐渐增高,同时B组和E组TNF-α和IL-6的水平明显低于A组,IL-2的水平明显高于A组,差异均有统计学意义(P<0.01)。结论蜂胶小分子凝胶对老年大鼠口腔溃疡有治疗作用,能明显缩短口腔溃疡愈合时间,治疗效果与蜂胶口腔膜相当,治疗机制与机体免疫功能调节有关,可能通过降低某些炎症因子如TNF-α和IL-6等因子和提高某些炎症因子如IL-2等因子水平使口腔溃疡愈合。
Objective To explore the effect and mechanism of propolis small molecule gel on oral ulcer in aged rats.Methods 50 male rats(12 months of age)were randomly divided into 5 groups with 10 rats each group.40% glacial acetic acid burning method was used to prepare a model of oral ulcer in aged rats.The model group(group A)had no drug treatment.Film group(group B)was given commercial propolis oral film from Zizhu Pharmaceutical,half a patch,once a day.Propolis micropowder group(group C)was given propolis extract micropowder 1 mg,once a day.Small molecule gel matrix group(group D)was given a small molecule gel matrix without propolis,half a branch,once a day.Propolis small molecule gel group(group E)was given propolis small molecule gel,half a branch,once a day.Rats in each group were anesthetized with 10% chloral hydrate,and the oral ulcer diameter was measured with an electronic vernier caliper before administration.Body weight was observed and the healing time was recorded.Interleukin(IL)-2,IL-6 and tumor necrosis factor(TNF)-α in serum were detected by enzyme-linked reaction adsorption method.Results Compared with group A,group B,group C and group E could significantly reduce the diameter of oral ulcers of rats,there were significant differences on the 3rd day of treatment(P<0.05).Compared with group A,the average healing time of group B and group E were significantly shortened(P<0.001).Compared with group A,TNF-α and IL-6 in serum of group B and group C at 1 d,3 d,and 7 d were significantly reduced and IL-2 in serum were significantly increased(P<0.01).Conclusions Propolis small molecule gel has a therapeutic effect on oral ulcers in elderly rats,which could significantly shorten the healing time of oral ulcers.The therapeutic effect is slightly better than that of propolis oral membranes.The treatment mechanism is related to the regulation of the body immune function.It might be through reducing certain inflammatory factors such as TNF-α and IL-6 and other inflammatory factors such as IL-2 to make oral ulcers heal.
