芪丹益肺通窍颗粒对变应性鼻炎大鼠血清IL-4、IFN-γ的影响

Effect of qidan yifei tongqiao granule on serum IL-4、IFN-γof allergic rhinitis rats

目的探讨芪丹益肺通窍颗粒对变应性鼻炎(AR)大鼠血清IL-4、IFN-γ表达的影响,初步探讨该方治疗AR的作用机制。方法将32只SPF级SD大鼠随机分为空白对照组、模型对照组、治疗组,治疗组又分为中药组与西药组,每组8只,雌雄各半。除空白对照组外,其余3组均采用卵白蛋白(OVA)腹腔注射致敏、双侧鼻腔滴鼻激发制备变应性鼻炎(AR)大鼠模型。造模成功后,中药组给予芪丹益肺通窍颗粒溶液灌胃,西药组给予氯雷他定混悬液灌胃,模型组灌胃等量生理盐水。连续给药14天后处死大鼠,取血清,观察一般情况及鼻部症状评分,计算大鼠主要脏器系数,采用ELISA法检测血清中白细胞介素4、干扰素γ含量。结果与空白对照组比较,模型组大鼠血清中IL-4水平明显升高、IFN-γ水平降低(均P<0.05);与模型组比较,芪丹益肺颗粒组和氯雷他定组证候表现明显改善,鼻部症状评分降低,血清中IL-4水平降低、IFN-γ水平升高(均P<0.05),但2个治疗组比较,差异无统计学意义(P>0.05)。结论芪丹益肺通窍颗粒可有效控制AR大鼠鼻痒、喷嚏等症状,其机制可能是下调IL-4水平与上升IFN-γ水平来纠正Th1/Th2失衡而对AR大鼠发挥治疗作用,减轻鼻部炎症反应,本研究为运用益气活血通窍法治疗变应性鼻炎提供了依据。

Objective To investigate the effect of Qidan Yifei Tongqiao Granule on serum IL-4、IFN-γexpression in rats with allergic rhinitis(AR),and to explore the mechanism of AR treatment.Methods 32 rats of SPF grade SD were randomly divided into blank control group,model control group and treatment group,The treatment group was divided into traditional Chinese medicine group and western medicine group,Eight in each group,Half male and half female.Except for the blank control group,The other three groups were sensitized by intraperitoneal injection of ovalbumin(OVA)and induced by bilateral nasal drip to prepare allergic rhinitis(AR)rat model.After making the model,The traditional Chinese medicine group was given Qidan Yifei Tongqiao granule solution to the stomach,The western medicine group was given loratadine suspension,The model group was given the same amount of normal saline.After 14 days of continuous administration,rats were killed,Take the serum,Observe general condition and nasal symptom score,Calculating the major organ coefficients of rats,The contents of interleukin 4(IL-4)and interferon in serum were detected by enzyme linked immunosorbent assay(ELISA)γ.Results Compared with the blank control group,The serum IL-4 level of rats in model group was significantly increased and IFN-γlevel was decreased(P<all 0.05);Compared to the model group,The syndromes of Qidan Yifei granule group and loratadine group improved obviously,Reduced nasal symptom score,Serum IL-4 levels decreased,IFN-γlevels increased(all P<0.05);and 0.05),But in two treatment groups,(P>0.05)The difference was not statistically significant 0.05).Conclusion Qidan Yifei Tongqiao Granule can effectively control the symptoms of itching and sneezing in AR rats,The mechanism may be to down-regulate the level of IL-4 and rising IFN-γto correct the imbalance of Th1/Th2 and play a therapeutic role in allergic rhinitis rats,Reducing nasal inflammation,This study provides an experimental basis for the treatment of allergic rhinitis by using Yiqi Huoxue Tongqiao method.
