摘要
以南极磷虾为原料,选取超声水洗时间、Ca(OH)2添加量、反应温度、反应时间进行单因素试验,利用正交试验法确定了南极磷虾酶解液脱氟的最佳工艺,即超声水洗时间20 min、Ca(OH)2添加量0.6%、反应温度40℃、反应时间60 min,此时酶解液氟含量为24.78 mg/kg;此外,分析了南极磷虾肽的分子量分布、氨基酸组成及抗氧化活性。结果表明:南极磷虾肽的分子质量小于1000 U的组分高达93.233%,氨基酸组成丰富,尤其富含谷氨酸、天冬氨酸及其酰胺;南极磷虾肽的抗氧化能力与其浓度呈现一定的依赖性,其清除DPPH·、·OH和O2^-·的IC50值分别是2.12,5.72和16.43 mg/mL,同时具有一定的还原力,说明南极磷虾肽具有良好的抗氧化能力,为南极磷虾肽应用于相应的保健食品、药品等领域奠定了理论和应用基础。
Using Antarctic krill as raw materials,the ultrasonic washing time,the addition amount of Ca(OH)2,the reaction temperature and the reaction time were selected for a single factor test.The orthogonal test method was used to determine the best process for defluorination of the Antarctic krill enzyme hydrolysate.That was ultrasonic washing time 20 min,the addition amount of Ca(OH)20.6%,reaction temperature 40°C and reaction time 60 min.The fluorine content of the enzymatic hydrolysis solution was 24.78 mg/kg;In addition,the molecular weight distribution,amino acid composition and antioxidant activity of Antarctic krill peptide were analyzed.The results showed that the components of Antarctic krill peptides with a molecular weight of less than 1000 U were as high as 93.233%,and the amino acid composition was rich,especially rich in glutamic acid and aspartic acid and their amides.The antioxidant capacity of Antarctic krill peptide had a certain dependence on its concentration.The IC50 values of DPPH·,·OH,and O2^-·were 2.12,5.72 and 16.43 mg/mL,respectively.They had a certain reducing power,indicating that Antarctic phosphorus shrimp peptide had good anti-oxidation ability,which laid a theoretical and application foundation for the application of Antarctic krill peptide in the fields of health food and medicine.
